This circular describes the activities and publications of the Bureau of Mines, Explosives Research Laboratory4 for the fiscal year 1960 (July 1, 1959 -June 30, 1960). Part 1 describes briefly the principal projects that were active during the report period, and presents some data not scheduled for publication elsewhere in the near future. Certain of the results are only preliminary and should be regarded as tentative, pending the findings of the completed investigation. Publications that appeared during the report period are listed and described briefly in part 2. In general, the research activities reported in these publications antedate fiscal year 1960. The published material relating to activities that are still current is indicated by references in part 1.

The Bureau of Mines is developing melting data for binary combinations of the alkaline earth fluorides and yttria to facilitate the formulation of slag composition. Liquidus and solidus temperatures and solid solubilities were determined for the BaF2-Y203 system, and the information is presented as the phase diagram. The phase diagram was constructed from data obtained by ceramography, X-ray diffraction, and thermal analysis of specimens encapsulated in small molybdenum ampoules. The data indicate a simple eutectic system with maximum terminal solid solubilities of less than 1 mole pct. The eutectic temperature is 1,343° C, and the composition of the eutectic is less than 1 mole pct Y203.

No minerals have been produced from the McGraw Creek RARE II area. During the 1982 U.S. Bureau of Mines study, there were no active mines in the area. Limestone occurrences (fig. 1, no. 2), four copper prospects (fig. 1, no. 1, 4, 5, 6), and a prospect with no significant mineral values (fig. 1, no. 3) were examined. A moderate potential for high-calcium limestone resources was estimated between ocurrences within the study area near McGraw and Spring Creeks (fig. 1, no. 2). There is low potential for copper resources at the Argenta Prospect (fig. 1., no. 1) in the study area, and the McDougall Prospect adjacent to the area.

The Bureau of Mines is developing methods for reclaiming the metal and mineral values contained in municipal incinerator residues. This report describes the first phase of the research, in which reliable methods for sampling and analyzing municipal residues were established and the composition and characteristics of the residues were determined. Samples were obtained from a variety of grate-type furnaces and also from a rotary kiln furnace. The samples were processed on a batch basis and separated into their natural categories by a variety of methods including screening, magnetic separation, air separation techniques, and handpicking. The main components of the residues were determined to be metal and metal oxides, glass, and ash.

Longwall minim; is a highly mechanized, high productivity mining method that is finding increasing applications in coal. Although the method would be highly advantageous in certain hard rock mining applications it cannot be applied in hard rock because no suitable cutterhead exists for such materials. The wedge longwall cutterhead is a, roller cutter device that is wedge-shaped, with cutters arranged to eliminate the very large normal force ordinarily required to cause such cutters to penetrate hard rock. However, the wedge geometry cannot be extended to a point: special nose cutters are needed to open a narrow slot for the wedge. This report describes the design, fabrication, and successful field testing of these critical nose cutters.

This Bureau of Mines publication is a literature study of the problem of spontaneous oxidation and combustion of sulfides in underground mines. It summarizes the present-day hazards, process mechanisms, and prevention and control techniques of this growing problem in underground mines. The iron sulfides generally have the greatest tendency to oxidize, particularly pyrrhotite. The presence of moisture and oxygen is essential for the oxidation reaction. Highly fractured sulfides, when found with timber or combustible refuse in slowly circulating air, pose the most serious fire hazard. Quenching with water is effective in quickly dealing with local hot spots, but ventilation at high flaw rates is the most effective and economical means for long-term prevention and control.

Although ASARCO Incorporated, the only domestic producer, had terminated copper smelting operations and associated recovery of byproduct arsenic trioxide at Tacoma, WA, in 1985, it shipped small quantities of both trioxide and metal from its remaining stocks in 1986. Imports for consumption of arsenic trioxide increased significantly, owing to increased demand for arsenical wood preservatives and the decrease in domestic shipments of trioxide. Prices remained unchanged throughout the year. Development of gallium arsenide integrated circuits, important in advanced military and commercial electronics circuitry, continued to gain momentum during 1986. Domestic Data Coverage.-Shipments of small amounts of arsenic trioxide and metal from remaining stocks by Asarco were reported voluntarily to the Bureau of Mines.

Toxic fumes generated during shot firing under actual mining conditions were measured. Eight shots with Gelodyn-2X and seven shots with Tovex-100 4VT were monitored. Average CO values for Gelodyn-2X and Tovex-100 4VT were 0.20 ft3/lb and 0.09 ft3/lb, respectively. Average N0x values for Gelodyn-2X and Tovex-100 4VT were 0.049 ft3/lb and 0.026 ft3/lb, respectively. The results obtained with these in-mine tests will be used by the U.S. Bureau of Mines in the development of a large chamber method of measuring fumes from shotfiring of explosives.

"This paper contains some new thermodynamic data for silicates in the series Mg2SiO4-Fe2siO4 and MgSiO3-FeSiO3 and gives the results of a thermodynamic attack on the problems of stability end parageneses.. Although some data are lacking satisfactory agreement with mineralogical and petrological observations is indicated.The use of thermodynamic methods in mineralogy and petrology has been rather limited, because accurate thermal data, especially for the more complicated and more important rock-forming minerals, have been too scanty to permit any systematic application to problems in those fields. It is, nevertheless, a fascinating task to investigate to what extent the mode of occurrence and the stability relations of the minerals and mineral parageneses, as revealed by petrological and mineralogical observations, agree with predictions of thermodynamics. At the present stage of development calorimetric data are needed for most of the abundant silicate minerals.Measurement of the heats of solution in hydrofluoric acid of minerals in the series Mg2SiO4-Fe2SiO4 and MgSiO3-FeSiO3 affords a ready means of obtaining their heats of formation from the oxides. Combination of such data with entropy and heat-capacity values gives a means of obtaining free energies of formation from the oxides, which, in turn, are connected di¬rectly with stability and paragenoses.The two silicates of magnesium, Mg2SiO4 and MgSi03, occur as rock-forming constituents as forsterite and clinoenstatite or enstatite. The orthosilicate (forsterite) forms a series of isomorphic mixtures with ferrous-orthosilicate (fayalite). The continuity of this series, suggested by petrological observations, has boon proved experimentally by Bowon and Shairer.4/ The present work shows that those minerals constitute a series of perfect solid solutions - that is, a series of solid solutions for which heats of formation from the individual pure silicates, Mg2Si04 and Fe2SiO4, aro negligibly small.The metasilicates of magnesium and iron also form an isomorphic series. However, this series differs from the forstorito-fayalite series in the fact that the most iron-rich mixtures, between about En10Fs90 and pure Fs, appear to be unstable with respect to the paragenesis iron-rich olivine plus quartz. The supposed instability of pure FeSiO3 as a crystalline compound, at least at high and at moderate temperatures, is remarkable, as no crystal struc¬tural reason for such instability is apparent. The present work indicates, also, that these minerals form a perfect series of solid solutions and throws additional light on the instability question.In the following, the samples used in this investigation will be described. For each of the natural olivines and orthopyroxenes, with the exception of sample L, a chemical analysis was made of the purified and actually used fraction. All the analytical work was carried cut by Oleg v. Knorring, and the specific gravity determinations, by moans of a pycnomctcr, by Oiva Joensuu, both of the University of Helsinki, Finland. The determinations of the optical properties were made by Mrs. Toini Mikkola, of the Geological Survey of Finland."

.1. The purification pilot plant and the equipment, solution, and gas cycle used for removing hydrogen sulfide and carbon dioxide are described. 2. As sulfur recovery is necessary in any large Fischer-Tropsch plant, pilot-plant runs were made at 300 p.s.i.g. to study the efficiency of tri-ethanolamine solutions in removing selectively hydrogen sulfide from gases containing varying amounts of carbon dioxide. 3. Aqueous solutions of TEA of 20, 30; 35, and 50 percent were investi¬gated. Data indicate that 30 to 35 percent is the optimum concentration if both selectivity and economy are considered. Slightly better selectivity is obtained with the.20 percent solution, but steam consumption is greater owing to the higher solution rates. Results from our pilot plant runs indicate that a 50 percent solution not only produces an acid gas of lower hydrogen sulfide content but also requires high solution rates to effect hydrogen sulfide purification. 4. Data from 37 runs using TEA are presented in tabular form. In addition, the data are correlated and presented in three sets of curves. By using these curves, it is possible to calculate the compositions of the purified and acid gases for various raw gas mixtures. 5. In cooperation with the Southern Natural Gas Co., investigations were carried out at 300 p.s.i.g. on the simultaneous removal of hydrogen sulfide and carbon dioxide, from gases using various concentrations of diethanolamine. 6. Data from 17 runs employing 20, 30, and 40 percent DEA are reported in both tabular and graphic form. 7. A comparison of the data from one DEA run made at 200 p.s.i.g. with those from the runs operating at 30O p.s.i.g. shows float the absorption capacity of the solution, and, hence, the steam requirements per gallon are about the sage for both pressures. .The only real advantage of the higher pressure is the additional volume of gas that can be treated. 8. The quantity of steam needed to reactivate fouled TEA and DEA solutions increased as the reactivator pressure was increased about 1,0 to 1.1 pounds of steam was required for each gallon of circulating solution when the reactivator was operated at 5 p.s.i.g. 9. A curve is included that shows the effect of the hydrogen sulfide con-centration in a lean 35 percent TEA solution on the hydrogen sulfide content of the purified gas and on steam consumption for reactivation. 10. Both TEA and DEA reduced the organic sulfur content of the raw synthesis gas considerably. Analyses of the lean amine solutions showed a gradual build up of sulfur compounds, indicating that the compounds formed by the amines and organic sulfur do not break down under the ordinary operating conditions of the reactivator. 11. Considerable operating difficulty was encountered in the pilot plant owing to foaming of the amine solutions. Laboratory experiments in which the effectiveness of certain antifoam agents was studied showed Alkaterge C to be most promising for the particular conditions studied.

"The Union Copper mine is in the Gold Hill district, Rowan and Cabarrus Counties, N. C.The mine was discovered in 1799 and first worked as a gold mine; but copper was encountered at depth, and the property became an important producer of copper. It is estimated that the mine produced 5,000,000 pounds of copper before operations were discontinued in 1907.As part of the strategic minerals program, the Bureau of Mines conducted a development project on the property from July 1943 to November 1943. Eight diamond-drill holes, totaling 2,500 linear feet, were drilled to delineate the ore body. Analyses of 205 core samples were made.Tabulations of sample analyses and drill-hole logs are presented in this report.The project work embodied in this report was under the immediate supervision of Austin B. Clayton, project engineer, and the general supervision of H. B. Ewoldt, district engineer for North Carolina and Tennessee. The preliminary report, written by Austin B. Clayton, was revised and the various figures redrawn for publication by T. J. Ballard under the general supervision of M. H. Kline, chief, Raleigh Branch, Mining Division.Acknowledgment is due the Metallurgical Branch for analytical determinations of the samples.The mine property is 1.5 miles southwest of the village of Gold. Hill, N. C. (fig. 1). Gold Hill is on U. S. Highway 52 between Salisbury and Albemarle and on a branch line of the Southern Railway. The latitude is 350 30' and longitude 80° 21 The tract consists of 553-1/2 acres in Cabarrus and Rowan Counties.A tentative list of the surface-rights ownership is as follows: William Hoy Barringer - 16 acres, A. W. Tucker - 1 acre, Frank Wagoner - 4 acres, and George Barringer - 3-1/2 acres, in Cabarrus County; A. W. Tucker - 2 acres, H. N. Hedrick - 54 acres, and William Atkins - 473 acres, in, Rowan County, N. C.The mineral rights are owned by Alfred Rossiter, Alice R. Hesslein? and the estate of the late Bella Rosenblatt. The attorneys for the owners are Neidle & Taylor, 6 East 45th Street, New York City.The Union Copper mine is in the Cold Hill mining district of North Carolina where gold was first discovered in 1799. The upper parts of the veins in the district were first worked for gold; however, the increase in the sulfide content of the veins with depth rendered the amalgamation process for gold recovery ineffective, and the mines were closed.The period of greatest activity at the Union Copper mine was from 1899 to 1906. At an earlier period the oxidized and leached part of the lode near the surface was mined from a working known as the ""big cut,"" the reported yield of which was $300,000 in gold.3/ The mine has not been operated since 1907.J. T. Pardee3/ lists the probable value of the production of the Union Copper mine as $1,050,000, of which about $675,000 was the value of some 5,000,000 pounds of copper produced."

Of all the war work by the Bureau of Mines none ultimately covered such a variety of processes and equipment, employed as many men, or called for as large an outlay as that started in con- nection with the solution of problems relating to the use of noxious gases in warfare. Beginning with an investigation to develop the best type of gas mask, the scope of the work extended until it included researches relating to a wide range of devices, such as different types of poisonous and irritating gases and smokes, smoke screens, gas shells and gas bombs, flame throwers, trench projectors for firing gas bombs, signal lights, and incendiary bombs. The Bureau of Mines in starting this work selected the necessary personnel, pro- cured the early equipment, and for some months, from the begin- ning to June 30, 1917, paid the cost of the work from its own funds. The Bureau of Mines built up a research staff of more than 700 chemists, including many of the most prominent chemists in the country, and obtained the cooperation of many of the universities and chemical companies. When the results obtained through this research were to be applied by the War Department, the Director of the Bureau of Mines cooperated in the selection of men to have charge of manufacture or further development, recommending chemiste who subsequently received commissions in the Army. The personal cooperation with the War Department was thus very close. The bureau can fairly claim that because it started this work on gas warfare and received the hearty support of the Department of War, the Navy Department, the National Research Council, and State, educational, and private institutions, the country was in July, 1918, months ahead of where it would otherwise have been in the production of gas masks and other devices. The production of toxic gases then far exceeded the supply of shells. In addition the later work in gas-mask manufacture by the Surgeon General's Office of the Army, which resulted in the development of the best mask produced anywhere; in gas manufacture and gas proving-ground tests by the Bureau of Ordnance, and in the chemical warfare pro- gram of the Navy, including the use of smoke screens, shells and toxic gases, were the direct results of the bureau's experimental work. The American charcoal and soda lime, developed through the research work, are the best used by any nation. The Bureau of Mines was instrumental in developing satisfactory methods of manu- facturing chlorpicrin, phosgene, mustard gas, brombenzylcyanide, cyanogen chloride, and other noxious substances on a large scale. The bureau also developed a smoke funnel for the Navy which no doubt saved many ships from submarine attacks, and devised smoke barrages, signal lights, incendiary darts, etc., for the Army. The Ordnance Department of the Army asked for incendiary bombs and were furnished with bombs better than any made previously. Perhaps the greatest single result of the research work has been the demonstration of the value of the services that the chemist and chemical engineer can render in time of need and the recogni- tion of chemical investigations as a necessary function of the Gov- ernment.

"The discovery of large deposits of polyhalite (K2S04.MgS04.2CaSO4.2H2O) in Texas has led to an investigation of possible industrial proecsses for the extraction of potash from this material. This mineral was first discovored in small quantities in the European potash deposits in or about the year 1800. Chemists fire thought it to be a form of calcium sulphate, but a more accurate analysis by Strohmiyer indicated the composition as given by the above formula.The decomposition of polytalite by aster is very slow and results in the formation of a layer of gypsum at the surface of the polyhalite which retarts the solution of the potaseium and magnesium sulphates. To obtain a substantial yield of K2S04 in solution it is necessary to use large amounts of water at room temperature, for he concentration of K2S04 is limited by the low solubility of syngonite (K2S04.CaS04.H2O) whereas at 100º C. several days boiling are necessary to obtain more than a 6 per cent K2S04 solution even when finely powdered polyhalite is employed.In 1854 H. Rose discovered that calcination, followed by leaching with water, resulted in a larger extraction of the potassium and magnesium sulphates, as compared with leaching without precalcination. During the course of a series of investigations concerning formation of oceanic salt deposits, Van't Hoff and his pupils obtained sufficient data to construct the equilibrium diagram for the system K+Mg+SO4--Ca++.H2O for 25° C. and for 83° C. These data are very well summarized. in the International Critical Tables' and by Boeke and Eitel. From a consideration of the diagrams for 83º C. one would conclude that an extraction of polyhalite with moderate amounts of water at 33º C. would result in a solution whose maximum concentration of K2So4 would be about 3.6 grams per 100 grams of water and of M2SO; about 4.0 grams per 100 grams of water, corresponding to equilibriumwith the solid phases: Polyhalite potassiam-calcium pentasulphate and anhydrite.The fact that it has been found possible to obtain as high as 11.4 grams of K2S04 per 100 grams of water (see table 6) and an equivalent amount of MgSo4 (8.1 grams per 100 grams of water was obtained corresponding to a ratio K2/Mg of 0.98; indicates that the formation of these complex salts is very slow. To be sure, this results was obtained at 100° C. instead of 83º C. However, the work of Anderson and Nestall 9 on the double salts of calcium and potassium sulphates at 100º C. indicates that the diagram at 100º C. C. would not be greatly different from that at 83º C. This slow rate of approach to equilibrium when combined with precalcination of,of polyhzli-,a is the 17,asis of some of the more p:olisinE methods of extracting potash from polyhalite."

Laboratory test results are presented for the determination of elastic modulus made with the NX borehole jack. Determinations were made in massive pieces of aluminum so as to satisfy conditions of linear elasticity, homogeneity, and isotropy. Results indicate a large discrepancy (approximately 200 percent) betwen material modulus as determined by laboratory block-loading tests and that indicated by the jack under relatively ideal circumstances. Effects due to interface friction, finite block size, incomplete plate-bore contact, and other factors were examined and found not to be significant. Finite element results indicate that assumed loading conditions for the jack differ from those actually occurring, at least in materials of high elastic modulus (approximately 10 million psi), such as quartzite.

Drive systems for mining machines are usually sized to accommodate anticipated peak loads. Usually, the average power requirements are considerably less. Energy storage and power averaging are attractive concepts for mining machinery and could have far-reaching implications. The purpose of this program was to: ? Identify truly feasible concepts, ? Identify advantages/disadvantages, limitations, and constraints, ? Evaluate feasibility based on today?s technology, ? Identify anticipated safety problems, and ? Provide the designer with a comprehensive literature base. Concepts evaluated include flywheels, high energy batteries, gas charged accumulators, inductive storage, capacitive storage, springs and counterweights, thermal, closed cycles, internal combustion engines, trolley systems, and hybrid systems. A comprehensive reference index is provided. Included is an appendix containing abstracts of the pertinent literature.

"The X-ray method of quantitative determination is advantageous because it permits determination of compounds rather -than ions or elements; for example, the direct determination of quartz in silicates,4/,5/,6/ The method also takes into account the finest particles of a sample, which may be lost in wet chemical methods because of solubility or in petrographic methods because of limitation of the microscope. The sample is usually reduced to a powder and is easily prepared,7/ only a few milligrams of material being necessary. The sample and its chemical composition are not destroyed and may be used for further analysis by X-ray diffraction or by other methods.This paper presents in detail some of the more essential problems that arise in the application of X-ray diffraction to quantitative chemical analysis, Photometric measurements are discussed in connection with line intensities and the choice of X-ray film. The internal standard method, successfully used in spectrographic analysis, is applied.Copper K-alpha X-rays from a demountable research-type X-ray tube were used with a camera of 20 centimeters radius with a cast-lead-slit collimating system. A combination of a tilted wedge and a lead-slit collimating system permitted irradiation of a large amount of sample with X-rays. This tended to minimize errors due to possible inhonogeneity of the sample, to decrease the time of exposure, and to keep the lines on the X-ray film to a minimum width. There was sufficient dispersion of the lines for satisfactory analysis of most samples. Power was furnished the tube by a. source of constant high potential consisting of a synchronous motor generator furnishing 500-cycle power to a 100-kv. transformer, rectifying valves, and a condenser filter circuit. The X-ray tube was operated as a gas-type tube at 35 kv, and 20 ma,, the exposure times varying from 1 to 2 hours. The constant output of X-rays from the tube, indicated bar an exposure meter built for the purpose, was satisfactory.The recording microphotometer8/has been described. It was found preferable to a nonrecording instrument for measuring blackening of lines above a background on X-ray films. This background, which usually occurs, is sometimes complicated by numerous peaks from lines of complex diffraction patterns. is microphotometer record of the pattern, therefore, facilitates the location of the background 'and the measurement of the heights of the peaks and thus contributes to the accuracy of a determination."

This work, conducted under Bureau of Mines contract, deals with the development of a computer graphics software package that will: 1. display mine ventilation schematics and other representative Illustrations for a real-time data acquisition system under development by the Bureau, and, 2. utilize the Bureau-owned Minefire and Ventilation Code for display of ventilation on and contamination predictions. The software package has been written try operate on a 64K, Z-80 based microprocessor-computer. The project has been divided into two phases. The first phase dealt with the development of the graphics software The sec rind phase dealt with the retrofit of the Minefire and Ventilation Code to the Z-80 microprocessor-computer.

"The premium on gold in London, reflecting the further depreciation of the pound sterling, has risen from about 22 per cent as noted in these reports for December, to about 28%. This will enable more South African gold mines to come into the profitably ;reducing class, and will also tend to increase the gold production from that region.Figures are now at hand showing the drop that occured in the sold output in the Transvaal, South Africa, during 1919, in which year 8,330,091 ounces of gold were produced against 8,418, 292 ounces in 1918, a decrease of 88,200 ounces. Annual gold output from this, the most important gold area in the world, has dropped almost one million ounces in the past three years, as the following table shows:Although the decrease in not as marked comparatively as that of 1918 over 1917, it is a further indication of the general world-wide curtailment of gold production, and the steady decline in production everywhere that is going on and will continue until some rational means is found to stimulate production, or until conditions return to normal.The subject of relief for the gold miner has received attention in all parts of the world, particularly in the United States and South Africa in the past few years, but in spite of all the plane that have been formulated to remedy the situation none have yet been adopted anywhere, due mainly to great danger of tampering with the gold standard. In the United. States a plan has advanced by H. N. Lawrie of the American Mining Congress to tax the new gold entering into manufacture of jewelry or the arts in general, as furnishing some measure of increased compensation to unprofitable gold mining.Gold production in Australia, as indication by figures issued by the Australian Mines department, shows a marked decrease, and for the first ten months of 1919 amounted to 914,957 ounces against 1,073,317 ounces for a similar period in 1918. Western Australia produces about two thirds of all the gold rained on that continent. The total gold production of Australia for 1919 will reflect the universal decrease in output.Ever since June 7, when the embargo on the export of gold Was lifted, there has been steady and heavy outflow of gold from this country, as a consequence of which considerable gold accumulated during the war has been exported. "INTRODUCTION Over the past several years a new type of cascade impactor that utilizes small nozzle diameters (micro-orifice nozzles) has been under development (Kuhlmey et al., 1981; Marple et al., 1981). The small nozzle dimensions allow for particles to be collected as small as 0.024 µm aerodynamic diameter on the last stage of the impactor. To obtain a reasonable flow rate of 30 I min - i through the impactor, a large number of nozzles (up to 2000) are required per stage.Furthermore, by locating the nozzles of multi-nozzle stages in a prescribed manner (described in detail by Marple et al., 1981) and rotating the impaction plates relative to the nozzle plates, uniform deposits can be achieved on the impaction plates. (1981), the advantages of a uniform deposit impactor over the conventional impactor that collects particles only beneath stationary nozzles are: (1) maximum use of the impaction substrate surface, reducing both particle bounce (if substrates are greased) and tare weights of the substrates (if particle mass is measured gravimetrically); (2) deposited particles can be analyzed for elemental composition by X-ray fluorescence (Dzubay and Rieke!, 1978); and (3) the pressure in the impactor can be kept relatively high compared to low pressure impactors that depend on a large slip correction to achieve small particle collection (some types of particles may evaporate at low pressures). The work described in this paper investigates two additional advantages of a uniform deposit impactor. These are: (1) the possibility of collecting two separate deposits at one time on one stage for two types of analysis techniques, and (2) the possibility of utilizing piezoelectric quartz crystal impaction plates more effectively by eliminating the deposit location sensitivity nature of the crystals (Schulze-Frohlich, 1981 ).MICRO-ORIFICE UNIFORM DEPOSIT IMPACTOR (MOUDI)A photograph of an eight-stage MOUDI is shown in Fig. 1. "The oxidation of NADPH by vanadate(V) in the presence of glutathione reductase showed typical enzymatic kinetics. The oxidation was inhibited by N-ethylmaleimide, a glutathione reductase inhibitor. Superoxide dismutase had no significant effect on the oxidation, indicating non-involvment of the superoxide radical. The vanadate(V) reduction was found to be a one-electron transfer process. These results suggest a new pathway for vanadate(V) metabolism and a new function of glutathione reductase. Compounds of vanadium(V), such as NaV03 , exert potent toxic effects on a wide variety of biological systems (1-3). Despite intense current activity, however, the biochemical mechanism of vanadate(V) toxicity is still not well understood (1- 14). It is believed that one of the important pathways involves the oxidation (by vanadate(V)) of NAD(P)H (15, 16), but, to our knowledge, there has been no report of the identification of any vanadate(V) reductase. With a view to finding if there exists any vanadate(V) reductase, we have investigated the oxidation of NADPH by vanadate(V) in the presence of glutathione reductase. Glutathione reductase (GSSG-R)2 was selected because our previous studies have shown (17) that GSSG-R can act as a reductase for chromate(VI), which is isoelectronic with vanadate(V). We find that GSSG-R does indeed function as a NADPH-dependent vanadate(V) reductase, thus pointing to a new pathway for the metabolism of vanadate(V), as well as to this property for GSSG-R. MATERIALS AND METHODSSodium metavanadate (NaV03 ), henceforth vanadate, was purchased from Aldrich and its solution was always freshly prepared. The GSSG·R used was from bovine intestinal mucosa and was purchased from Sigma. The oxidation of NADPH was followed photometrically as the decrease in absorbance at 340 nm. The spin trap, 5,5-dimethyl- pyrroline-N-oxide (DMPO), was purchased from Aldrich and used without further purification, since very weak or no spin adduct signal was obtained from the purchased sample when used alone.ESR spectra were obtained at X-band (9.7 GHz) using a Bruker ER200 ESR spectrometer. The magnetic field was calibrated with a self-tracking NMR gaussmeter (Bruker, Model ER035A) and the microwave frequency was measured with a digital frequency counter (Hewlett-Packard, Model 5340A). An ASPECT 2000 computer was used for data storage and analysis. The concentrations given in the figure legends are the final concentrations. All experiments were carried out in the phosphate buffer solution (pH 7.2) and at room temperature."
