And a new generation of safety culture experts are telling mining companies how to use it: It is a situation that is still too common in the mining industry: the latest personal protective gear is in place. The safety rules are known and followed. And then, in a moment of inattention or carelessness, a mistake is made and someone gets hurt or killed. Common wisdom claims that such accidents are impossible to eliminate entirely. Mining is dangerous, and some miners will always be hurt and killed. An increasing number of mine safety experts, however, reject the common wisdom. A year with zero mining deaths is possible, they say; with the use of one of the newest weapons to the mining industry in the safety war: the concept of safety culture. Safety culture refers to the set of attitudes, beliefs, perceptions and values that influence safety in the work environment. It is also sometimes called the way an organization behaves when no one is watching. Robert Peters, of the Office of Mine Safety and Health Research (OMSHR) at the National Institute for Occupational Safety and Health (NIOSH), describes safety culture this way: ?It?s what?s believed on a day-to-day basis. For example, if a miner sees a workplace hazard and he truly believes that his boss wants him to identify the hazard and bring it to the company?s attention, that would be an example of a positive safety culture.?

Throughout the 1970s, the US Bureau of Mines rode an ebb tide in Washington political circles. In rapid fire succession, the agency lost its mine health and safety enforcement responsibility (1973), a large portion of its energy research responsibility (1975), and then its fuel statistics functions plus a sizable portion of its coal R&D (1977). By 1978, johnny-come-latelies in the Energy, Labor, and Interior Departments had siphoned away from the Bureau some $125 million in program responsibilities as well as 900 of its employees. The bruising decade came to a close with a Department of Natural Resources proposal that, in its original form, would have scattered the Bureau's responsibilities to the winds. "There is no doubt in my wind," said Lindsay Norman, acting director of the Bureau of Mines (see box), "that were the Bureau to continue down the path it was on, the next step was closing the doors and going out of business." The doors are still open, but erosion of responsibilities had a devastating effect on the resources available to fulfill the Bureau's mission. Devastating, too, in terms of internal morale, considering the Bureau had six directors in a 10-year span. "When the troops are in the trenches and are being shot at," Norman said, "they need a leader-hopefully, a permanent one- that can get them advancing. This is something the Bureau did not consistently have in the 1970s." In September 1978, Roger Markle climbed aboard as director of the Bureau to rejuvenate the 70-year-old agency. He resigned only six months later* but the program Markle set in motion is being carried through by Lindsay Norman. Since Norman's appointment as acting director, he has spent considerable time on the road explaining his agency's redefined goals. At one such stop in Duluth, Norman and his two deputy directors, Charles Kenahan and Mike Kaas, answered the following questions. Lindsay, you were the principal architect in the Bureau's 1979 reorganization. How about a glimpse of what that entailed? Toward the end of the 1970s, after almost a decade of steady dismemberment, we reached what we felt was a critical point in the Bureau's history-we had the choice of redefining and strengthening our organization to meet the mineral challenges ahead, or continuing down the path toward the agency's eventual demise. We felt the Bureau's mission too important to abandon, so in 1978 with Roger Markle as director we initiated a six-month, no-holds-barred intro spection of just what the Bureau should be doing, why, and toward what ultimate national needs. And what did this self-reflection reveal? That there is, indeed, a unique federal role for the Bureau of Mines-that of a lead agency for influencing mineral decisions and pioneering tomorrow's technologic advances. Don't confuse us with policymakers, we're not. But we are policy analysts; we're scientists, engineers, and mineral professionals who can offer a sorely needed source of objective, accurate information to decision makers in both the public and private sectors. I feel there is no better brain trust in government than the Bureau when it comes to mineral expertise-of course, I'm biased. Do I understand you to say the Bureau of Mines is an advocate agency for the minerals industry? We plead no parochial interests, nor ought we, but from our viewpoint a sound minerals industry is extremely important to the broader national goals of security and a healthy economy. US dependence on foreign sources for critical minerals, the increasing noncompetitiveness of some US mineral sectors, and the array of land use issues surrounding mineral-rich lands all have much more serious implications than many otherwise informed people seem to recognize. Two major types of economic secondary iron ores occur in the banded iron formation (BIF)-rich Hamersley Province of the Pilbara, Western Australia:dominant BIF-hosted bedded iron deposits (BID)detrital ore deposits (DID sensu lato), which are mainly hosted in the three province-wide Cenozoic sequences that include coeval non-ore sediments and comprise:a. the major Miocene, fluvial channel iron deposits (CID)b. the lesser Eocene and Pliocene detrital iron deposits (DID sensu stricto).Striking differences exist between the massive CID resources and the much smaller underlying and overlying DID. CID are essentially riverine alluvial ooidal rocks with abundant small fossil wood fragments and variable iron peloids, but with only extremely rare recognisable lithic remnants. The original matrix is typically ramifying layered goethite. Eocene DID are mainly alluvial with minor pisoids, whereas Pliocene–Quaternary DID are dominantly coarse colluvial gravels, both derived from and largely retaining the original textures of BID, hard cap or variably ferruginised surface BIF. The colluvial DID matrix is typically ferroan-aluminous soil, resulting in canga where replaced by goethite, which may be dehydrated to hematite by exposure. The Cenozoic deposits occur in two dominant geomorphological environments: the southern Marra Mamba to Brockman Iron Formation strike valleys containing all three Cenozoic sequences and the much later northern Brockman Iron Formation plateau valleys that include only the Miocene and Pliocene sequences. The Cenozoic detritals formed in different climatic regimes, with an extended dry period forming a prominent province-wide dehydrated carapace on the Eocene DID. The Miocene ‘optimum’ followed with its thick scrub-covered deep regolith that produced the fossil wood-rich CID and was succeeded by the arid cool period of the Oakover limestone/calcrete. A major renewal of exposure and erosion in the Pliocene resulted in the extensive iron gravels of the Pliocene–Quaternary.CITATION:Morris, R C, 2015. Abstract - Cobalt, Ontario, is renowned for the 12.6 billion grams (445 million ounces) of silver produced from the area since discovery in 1903 by workers of the Timiskaming and Northern Ontario Railway. Native silver generally occurs with cobalt arsenides and sulfosalts in near-vertical carbonate veins cutting the Huronian sedimentary rocks of the Gowganda Formation, the Archean metavolcanics and/or the Nipissing diabase. All major deposits have been found within a few hundred meters of the unconformity between the Archean and Huronian rocks in general proximity to the Nipissing diabase and volcanogenic sulfide mounds within the Archean meta-volcanics. Silver has been mobilized from one or more of the local country rocks by hyper-saline brines and deposited in or near zones of mixing where the saline brines encounter paleometeoric water transported to depth along the unconformity or local structures. Previous work has shown that chloride complexes are the dominant ligands responsible for silver transport. These hypersaline brines, represented as halite-bearing fluid inclusions at room temperature, have been trapped as primary fluid inclusions within vein minerals. Pressure-temperature conditions of vein formation have been derived from mineral equilibria, maximum lithostat and fluid-inclusion studies. These data are consistent with vein formation occurring over the temperature range 300°C to 350°C, with pressures constrained between 60 Mpa and 136 Mpa (600 bars and 1360 bars).

FEW men reach middle life without having had the experience of failure in one or more undertakings; and most of us can look back with gratitude to help or advice given us by friends at critical periods of our lives. If, as men, we ask and take the benefit of such aid, why should we expect our sons to be stronger? A parent can make no greater mistake than to send a boy, however steady and intelligent he may be, off' to school or college, and expect the boy always to cope successfully with all the new conditions and new difficulties that may confront him. This is undoubtedly more the case with a freshman, in his first year at college, than in later years, or than it is with a boy at day- or boarding-school, where the boy receives a greater degree of care and oversight than can be given to the young man entering college; of all periods in life, the youth in his freshman year-fresh from the overshadowing care of home and school-may need judicious advice and timely aid. The change from the restraint of home and school to the freedom of college life is sudden and marked, and it is in this the first year of the boy or young man of 17, 18, or 19 years of age, that some tactful, judicious aid should be near him in his work and course. Some boys go to college so well prepared that they become careless and finally fall behind in their work from over-confidence. Others meet trouble from ambitiously trying to do more than their previous preparation, or physical or mental strength, can sustain. Others fall behind from illness-from injuries-from natural inability to grasp at first, without more aid than comes to them in the regular course of instruction, the higher mathematics-or mayhap the modern languages, or other subjects presented by the advanced curriculum of the present day. PRACTICALLY all underground work on the Minnesota iron ranges is done by miners working on a so-called contract wage system. This system, while it has certain advantages over the straight day's pay or company-account method of wage payment, has serious and vital defects. In view of the scarcity and demands of labor, it is important that employers of labor scrutinize their systems of wage payments carefully, not only to save losses and trouble, due to labor disturbances, but also to secure the greatest efficiency of their workmen and reduce their operating costs. It is admitted that an adequate, just, and equitable wage is the prime factor in bringing about these benefits. The wage of labor and its method of application and payment have been, and always will be, the main factors in the unrest and dissatisfaction of labor. Not only does labor want and need an adequate wage, assuming that work is adequately performed, but it wants and is entitled to a just and equitable wage. Labor must have respect for the fairness of the wage and for the fairness of the management in the application of the wage. It is conceded that the Minnesota iron miners are now, and have been, in the main paid adequately, but the inherent defects of the so-called contract system prevent any just or equitable application of this wage to the various units of the labor body. The defects of the contract system, with its obvious inequalities, favoritisms, and injustices, are cancers in the brain of labor. Every thinking miner realizes these defects and knows just what they are; the unthinking do not realize the details of the subject, but each month they are forcibly reminded of these defects through the pay envelope. As practiced on the iron ranges, however, the so-called." contract" system is not a contract and the name is a misnomer. No true contractual obligation exists.

"Two in vitro systems are being developed to model initial stages in the interaction of respired dusts with the acinar region of the lung. Incubation of quartz and kaolin with dipalmitoyl glycerophosphorylcholine, a major component of pulmonary surfactant, suppresses the prompt C)1otoxicity of both dusts as measured by in vitro enzyme release from pulmonary macrophages or assay by erythrocyte hemolysis. Additional data is presented to quantitate the detoxification of quartz and kaolin by dipalmitoyl lecithin. Subsequent incubation of lecithin treated dusts with phospholipase enzymes results in partial to complete restoration of the assayed cytotoxic potentials, dependent on the lipase activity applied, the incubation time, and the mineral. Restored cytotoxic potential is due to removal of the prophylactic lecithin surface coating and/or due to residual lysolecithin which is produced by the partial enzymatic digestion of lecithin. Data is presented on the contribution of each mechanism to the restored toxicity. Again, this is dependent on the mineral used, the lipase activity applied, and digestion time. Additionally, a system is under development to challenge pulmonary macrophages in culture with native and surfactant treated dusts. A preliminary test of the dipalmitoyl lecithin - quartz rabbit pulmonary macrophage system is presented to illustrate the method.IntroductionRespirable quartz dust exposure is known to cause librotic lung disease; respirable clay dust exposure is less likely to do so as indicated by epidemiologic data on occupational exposures and by animal model studiesP·7) The hypothesis that damage to pulmonary macrophages in the acinar region of the lung by respired dusts is an early event in the disease process requires consideration because kaolin clay and quartz dusts are comparably cytotoxic to pulmonary macrophages in vitro.(3•9)"

Men and institutions are what they are largely because of inheritance. Out of the past comes experience that enables men to build more wisely than did preceding generations. The field of possibilities has widened as the horizon of men's vision has been extended. The realization of these possibilities has depended largely on the application of men's natural endowments, brain and brawn, to the problems at hand. In the field of mechanized mining, Wyoming has a greater percentage of men with vision, who have a realization of the possibilities of mechanized mining, than any other state in proportion to tonnage production. With the introduction of mechanized mining systems have come problems that have caused the men who are connected with the safety work much thought. Discussions on mechanical drilling, cutting, loading, conveying, haulage, etc., have come in for their share of the debate. There have been a few attempts made to show that mechanical mining is safer than are the more primitive ways of doing underground work by man power. We also have a number of places in the coal producing States of the United States, where the mechanized systems have been abandoned chiefly on account of the safety element involved bringing about condemnation of the system by State inspection forces. Certain types of scraper systems have been condemned, and some have voiced opposition to long faces. This would lead us to believe that the question is still a debatable one as far as the safety element is concerned. It is only fair that we discuss the operations of mechanical systems and from the results obtained take analysis of conditions as they exist, basing our conclusions from the results obtained.

If the problem be one of science or of industry we turn to the library of the subject and endeavor to translate the present from the experience of the past. I wonder if you would bear with me for a few minutes as I look back to the time when I was going through mining school and looking forward with hope, it is true and with confidence also, but still with some trepidation and an occasional qualm, to the month of June when the class of 1897 would be hunting jobs. Those were the years 1893 to 1897. Those years saw the Treasury of the United States so empty of gold that the President had to appeal to financiers to accept the country's promises to pay in return for $100,000,000 in gold. And the invectives that were hurled at both their heads for such a betrayal of the people sound strangely similar to those you read today being daily written and screamed at another President for taking us into the League of Nations or for not issuing bonds in huge amounts for unemployment relief, or for not adopting somebody else's particular specific for the relief of hard times.

This paper describes how Automated Mining Systems (AMS) is evolving the state-of-the-art in guidance systems for autonomous underground vehicles. The techniques, trade-offs, and lessons learned from these systems are described, including the safety regime that has been implemented. First generation autonomous guidance systems for underground vehicles relied on infrastructure for the vehicle to follow. However, there are additional costs associated with that approach. The current generation uses a pair of laser range finders as its only guidance sensors. This system uses a minimum of infrastructure, in the form of a redundant system of location referencing. Advanced steering and drive control algorithms are incorporated into this generation of system. The next generation of infrastructureless guidance systems will be based on a 3D vision sensor, which was originally developed for autonomous Mars rovers and is being adapted for underground mining use. Results of early underground field trials are presented, along with a description of this emerging technology.

Fatigue in mining operations is a serious issue and a significant contributor to incidents and accidents. While mine operators are using or introducing new technology to monitor operator fatigue, there is little work that focuses on the subjective experience of fatigue and the use of fatigue monitoring technology. To investigate this issue, eight focus groups with a total of 44 operators of heavy haulage equipment working for two mining companies in the Mountain West of the USA participated in 1-h focus groups. The topic of the focus groups was the experience of operator fatigue and issues surrounding fatigue at the workplace. The results of the content analyses suggest that operators are well aware of the signs of fatigue, and that they understand the severity of these signs as well. In addition, they are aware of the effect of fatigue on driving performance and are using a wide range of methods to manage fatigue. Finally, based on participants’ assessment, current fatigue monitoring systems lack face validity and reliability and are perceived as not being effective. The results of this study emphasize the importance of a socio-technical systems perspective in the context of miner health and safety management. Only such a comprehensive perspective can result in improvements that are sustainable.

"The Fort Knox gold deposit is hosted within the Fort Knox Pluton, a Cretaceous multiphase granitic body that intrudes the Fairbanks schist. Gold occurs along the margins of stockwork veins and veinlets, quartz-filled shear zones, and along fractures within the granite. The mineralization is generally sulphide-poor and closely associated with trace amounts of bismuth, tellurium and to a lesser extent with molybdenum and tungsten. At present, proven/probable reserve estimates, based on a cutoff grade of 0.39 g/t Au, are 158.3 million tonnes grading 0.83 g/t Au.The Fairbanks schist comprises the bulk of the country rock. The dominant lithologies present include muscovite-quartz schist and micaceous quartzite. Immediately north and south of the main orebody, mixed metamorphic lithologies of the Cleary sequence, a subunit of the Fairbanks schist, have been mapped. Development of local zones of weakness, most likely due to a broad regional warping event, resulted in brittle-ductile deformation and activation of a northwest-southeast shear couple between contrasting lithologies.The Fort Knox Pluton consists of at least three phases based primarily on textural differences. Intrusion of a fine-grained granite was followed by a medium-grained porphyritic granite. The youngest intrusive phase is a coarse-grained, seriate porphyritic granite, the Vogt Stock. The Vogt Stock forms an elongate dome with the axis in the east-west direction. Structures displayed in the overlying units, believed to be the result of doming, include tension fiSSUres and longitudinal fractures.Premineralization fractures, the result of the magmatic doming, provided conduits for initial stockwork quartz vein development and quartz enrichment in shear zones. Weak development of vein-controlled phyl/ic, potassic, albitic, and argillic alteration styles are present. Evidence of a late thermal event resulting in a low-temperature alteration assemblage of calcite + zeolite (stilbite) + chalcedony + clay occupies breccia zones, joint surfaces, and fractures within the granite."

TILNEY, the great expert on the study of the development of the brain of human beings and animals, tells us that the greatest difference between the human brain and the brain of animals is that man alone has shown evidence of being able to predict the future. Just why he throws overboard the busy bee who lays up honey for the winter, the squirrel who gathers nuts, and the humble ant, I do not know. At any rate, it is nice to be assured by an authority that we have this power of prophecy. My own experience with attempting to prophesy the future has convinced me that our only possibility of being good prophets is to be good students of history.

Ladies and Gentlemen: As we begin these three crowded days of technical and economic discussion of problems faced by mining and metallurgical engineers, it seems appropriate at this Keynote Session to consider the future of this industry from which we derive our livelihood, It is also appropriate at this international meeting to view our industry on a world-wide basis. First, let us assess the world's future needs for minerals - the needs which it is our industry's task to meet. Next, let us look at what supplies of minerals may be available to meet those needs. I will speak of this first in terms of the physical availabilities of minerals. Finally, we can consider some of the social or institutional factors which will condition the world's mining industry as it goes about its task of discovering, developing, producing, and delivering the minerals needed to meet the world's demands. Let me give you a foretaste of my conclusions. First, future world demands upon our industry will be enormous, eclipsing anything that we have known up to this point. Second, there is strong evidence that our earth's crust contains a sufficient physical quantity and variety of minerals to meet forthcoming needs for as long a period ahead as one can envisage.

Gold mineralization at Kidston occurs in a semicontinuous zone near the margin of a large, late Palaeozoic breccia pipe. The pipe is ovoid in plan, funnel-shaped in cross section and straddles a contact between Proterozoic metamorphic and granitoid rocks which are intruded by a swarm of late Palaeozoic acid dykes. Rock fragments within the breccia pipe reflect the enclosing country rocks and mix in various proportions to form gradational lithological domains. Most of the gold is free and is located in the breccia matrix and in a set of inwardly dipping sheeted quartz veins which kink around the pipe margin. Sulphide minerals, dominantly pyrite, occur as disseminations in the breccia matrix and as aggregates in the cores of quartz veins. Hydrothermal altLration is pervasive within the breccia and is most intense in areas with the highest percentage of acid dyke fragments. Sericite and carbonate are the main alteration minerals. An empirically developed genetic theory for the formation of the breccia pipe and attendant mineralization involves magmatic and post-magmatic processes associated with the intrusion of a granitoid body to a shallow level in the crust. Economic evaluation of the deposit relied heavily on the results of diamond drilling. Both polygonal and geostatistical reserves were calculated and found to be in close agreement. Geostatistical reserves are considered to more closely reflect the tonnes and grade achievable during mining. "Porphyry molybdenum-tungsten mineralization on the east flank of Hudson Bay Mountain crops out over 5 km2 and is intersected to depths of 2 km. Features such as a hornblende-biotite aureole, mineralogical zoning and dike, vein and fracture geometries within lithologies of the Middle to Lower Jurassic Hazelton Group and Cretaceous Skeena Group reflect an intrusive complex, the Upper Cretaceous to Early Tertiary Hudson Bay Mountain stock which has been intersected by drill holes. The stock is believed to be the mineralizing agent. A southeast-dipping granodiorite sill has been outlined by drilling within the Hazelton Group. Molybdenite and scheelite veins cut all rock types; however, the granodiorite sill hosts a higher density of fine-grained, banded, quartz-molybdenite and pegmatitic quartz-molybdenite veins reflecting a competency contrast between the intrusive sill and other country rock. Due to the relative abundance of these veins, grade is higher within areas of the sill. Above the 15000£ cross-cut 20.6 million tonnes grading 0.401% MoS2 and 0.041% W03 have been defined. During exploration of the deposit sampling procedures had to be adjusted because of the nugget effect attributed to drill intersections of banded and pegmatitic veins.IntroductionMolybdenite was discovered in outcrop at the foot of Kathlyn Glacier on Hudson Bay Mountain and first reported by the Geological Survey of Canada in 1944. The deposit has several names: Hudson Bay Mountain or Glacier Gulch, both after its location, and Yorke-Hardy after the original staker.The deposit is on the east flank of Hudson Bay Mountain, 9 km northwest of Smithers, west-central British Columbia, within NTS area 93L/14 (54° 49' North, 127° 18' West, Fig. 1). Hudson Bay Mountain, on the southern edge of the Bulkley Valley, is a dominant topographical feature reaching 2590 m elevation. Mineralization is partly overlain by the Kathyln Glacier which is visible from Highway 16. Access to the 3100 m of underground development is from a portal at 1066 m elevation on the east slope of Hudson Bay Mountain."

"Electron spin resonance (ESR) spin trapping methodology has been used to study the reactions of manganese dusts with glutathione, employing a-{4-pyridyl-1-oxide)·N-tert-butyl-nitrone (PBN) and 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) as spin traps. The results show evidence for the glutathionyl radical formation in the reactions of both MnO2 and manganese (Ill) acetate with glutathione. Based on these experimental observations, we hypothesise that the mechanism of neurotoxic effects of manganese might be due to the fact that the generation of glutathionyl radicals depletes the glutathione pool and reduces the glutathione shield against free radical products of dopamine metabolism and manganese induced reactive oxygenated species. IntroductionExposure to manganese compounds often occurs as the result of foundries and mining operations (1. 2). Other significant sources of environmental manganese include coal-fired power plants and burning of residual and crude oil (3, 4). Lately gasoline combustion has also started to contribute since manganese (methyl cyclopentadienyl manganese tricarbonyl. 0. 125 g/gallon) is being used instead of lead as an antiknock ingredient in automobile fuels (5. 6). Although manganese is an essential element in certain biological processes it is toxic in excessive amounts. The manganese toxicity is mainly associated with the nervous system (7-9). Symptoms frequently observed in manganese poisoning or manganism are disorientation, memory loss, anxiety, compulsive laughing and crying, mask-like faces, akincsia rigidity and tremor (10). In spite of numerous investigations to understand the mechanism of manganese induced injury. it has not been possible to localize the initial chemical lesion responsible for altered physiology and ultimate cell death. This may perhaps he the reason for failure in developing a successful therapy for manganese intoxication."

Abstract - The Horne mine was truly a world class Cu and Au deposit. Between 1927 and 1989, it produced some 260 t of Au and 1.13 Mt of Cu from 53.7 Mt of ore that averaged 2.22% Cu, 6.1 g/t Au and 13 g/t Ag. The total value of Au and Cu production from the Horne deposit at metal prices of US$300/oz Au and US$1.00/lb Cu is an outstanding US$5.2 billion. The Horne mine was also a company builder. After optioning the property from Ed Horne (Tremoy Syndicate) in 1922, the Thomson-Chadbourne Syndicate discovered the deposit in 1923 and quickly grew to become Noranda, one of the world’s premier mining companies. This discovery fuelled exploration and, along with subsequent discoveries in the Val d’Or-Cadillac camps, led to the "economic development" of northwestern Quebec. The Horne deposit influenced and continues to influence genetic models for volcanogenic massive sulfide (VMS) deposits. Early observations at Horne contributed to an epigenetic replacement theory for VMS deposits. The most recent genetic model for the Horne, invoking sub-seafloor sulfide replacement of silicified and sericitized volcaniclastic host rocks within a graben, has subsequently been proposed for another giant VMS deposit, the Kidd Creek mine. The Horne and Kidd Creek deposits show many similarities, such as localization within synvolcanic grabens, long-lived hydrothermal activity uninterrupted by volcanism, sub-seafloor replacement sulfides, stacked sulfide lenses, zone refining, silicified footwall rocks characterized by high positive ?18O values, and association with FIII rhyolites. Notable differences between the two deposits include the lack of andesitic, basaltic or komatiitic flows at Horne, different inferred water depths, high Au content at Horne versus negligible Au, but sub-economic to economic concentrations of Sn, In and Cd at Kidd Creek.

The inhalation of silica has been shown to produce a dramatic inflammatory and toxic response within the lungs of humans and· laboratory animals. Currently, no effective treatment exists for workers who may have been exposed to the inhalation of silica. The objective of this study was to develop an animal model in which we could evaluate the effect that anti-inflammatory steroids have on the acute silica-induced pulmonary inflammatory response. Male Fischer 344 rats were pretreated with either dexamethasone (2 mg/kg) or saline vehicle (i.p.) on days 1, 3, and 5. On day 6, the animals from the two groups were then intratracheally instilled with either silica (20 mg/0.5 ml saline vehicle) or saline vehicle (0.5 ml). Twenty-four hours after the instillations in the non-steroid group, significant increases occurred in total protein, total number of cells, neutrophils, and lymphocytes recovered from the lungs of animals treated with silica compared to saline controls. Silica also caused dramatic increases in the luminol-dependent chemiluminescence (LDCL) of lung tissue and bronchoalveolar lavage (BAL) cells. The LDCL reaction was markedly decreased by either superoxide dismutase (SOD) or N-nitro-L-arginine methyl ester hydrochloride (L-NAME). SOD is involved in the enzymatic breakdown of superoxide anion, while L-NAME, a nitric oxide (NO) synthase inhibitor, prevents the formation of NO. When the superoxide anion and NO react, they form the highly oxidizing substance peroxynitrite. This study then implicates peroxynitrite as an agent which may be involved in the silica-induced oxidant lung injury. When the animals were pretreated with the steroid dexamethasone, there was a complete protection against the biochemical, cellular, and chemiluminescent indices of damage caused by silica. The mechanism in which the steroid protects the lung from damage may be due to the ability of dexamethasone to block the induction of NO synthase. With further study in animals, the anti-inflammatory steroids may be useful in the treatment of silicainduced lung injury.
