Blockchains are one of the prominent disruptive technologies that are being implemented in several disciplines. The application of blockchains in the mining and mineral industry can provide utility for several processes especially when the industry is undertaking digital transformation. The governing principles of blockchain technology are applicable in undertaking transformation in several processes within the mining industry. This study investigates the applications of blockchain technology within the realms of the mining industry and identifies key areas of implementation. A systematic review of publications on academic platforms relating to blockchain technology in the mining industry did not retrieve a considerable number of articles. Consequently, a thorough review of white papers and articles authored by individuals on use cases within the mining industry is compiled and presented in this manuscript. This paper identifies all the potential areas within the mining industry where blockchains could provide value and improve processes through their implementation. The broad area of implementation of blockchains includes supply chain, traceability in spare parts, contract management, regulatory compliance, and cyber security.

The rock quality designation (RQD) is a commonly used index for the classification and evaluation of rock mass quality and is widely adopted in mining and geological engineering. The traditional method of obtaining RQD still requires manual measurement of core length and calculation of RQD, which is inefficient. To address this problem, we propose a framework for drilling core image segmentation and RQD estimation from digital images of cores in core boxes based on the deep learning algorithm. The proposed framework is generated by combining the Mask Region-Based Convolutional Neural Networks (Mask R-CNN) instance segmentation algorithm, the U-Shaped Convolutional Neural Networks (U-Net) semantic segmentation algorithm, and the image processing functions of the Open Source Computer Vision Library (OpenCV). To demonstrate the accuracy of the proposed method, seven boreholes in the Xiushuihe vanadium-titanium magnetite mine, located in the Sichuan Province, China, were used as the case study. According to the comparison of the manual measurements and calculations of the cores taken from seven boreholes used to conduct this study, the framework can record the length of the drill cores and calculate RQD within an average error rate of 3.42%, while it saves about 85% of the working time. The results illustrate that the proposed framework enhances the efficiency of RQD calculation while satisfying the engineering requirements.

"Pressure oxidation (POX) leaching of copper (Cu) concentrates continues to be explored as the next generation of leaching processes to sustain existing Cu Solvent Extraction/Electrowinning (SX/EW) operations as easily leachable ores become scarcer. The leach liquors produced, particularly from total pressure oxidation, contain elevated concentrations of Cu and acid, making solvent extraction processing more challenging. Addressing this challenge will require innovative processing techniques and continuous improvement to be successful in achieving satisfactory physical and metallurgical performance in the SX plant. The ability to efficiently process these feeds via SX plays an important role in the continued success of POX technology. Characteristics of a concentrate leach pregnant leach solution (PLS) are discussed, along with best practices that should be considered for processing by SX. Findings discussed in this paper will shed light on ways to reduce process risk and bring additional improvement to the industry with minimal change to the SX plant. INTRODUCTION Hydrometallurgical processing of copper concentrates continues to gain interest from the industry. Treatment by smelting and electro refining continues to dominate, accounting for ~80% of global copper production, but is faced with its own challenges. The case for increased hydrometallurgical processing has been discussed as high grade ore deposits are diminishing, capital costs for smelting are high, and the environmental aspects present challenges for sustainable practice. The hydromet copper industry is also turning a corner as easily leachable copper oxide deposits are depleting, and excess SX-EW capacity exists in certain regions. The transition to hydrometallurgical processing of copper concentrates will take a bit of effort as leaching of copper sulfides is difficult and byproduct credits must be considered. Various POX leaching processes have been studied and proposed to the industry as viable practices. Of the many processes, commercial use has been practiced with the Mount Gordon Copper Process, total and partial pressure oxidation by Freeport-McMoRan at their Bagdad and Morenci, Arizona complexes, the Sepon Copper Process in Laos, and the bioleach BIOCOP™ process in Chile. The experience related to the current paper centers around Cu concentrate leaching performed by Freeport McMoRan (known as CLP). The patented process was developed as an alternative to smelting and refining in 1998. The technology applies to both molybdenum and copper concentrates. Commercial CLP research and development began with Phelps Dodge in 1999 to 2001. The first commercial plant was commissioned in 2003 at the Bagdad operation. Initial operating conditions of the Bagdad CLP were under high temperature conditions before converting in 2005 to medium temperature conditions. In 2007-2009, the CLP technology was commissioned in Morenci, operating medium temperature and high temperature lite conditions until shutting down due to the economic downturn. The Morenci CLP restarted operation in April 2015 due to acid production benefits for the Morenci leach operations. Renewed solvent extraction test work initiated well before restart and has been an ongoing focus for further SX plant optimization."

Significant advances in longwall mining technology and equipment have occurred over the last decade. By the late 1990s, longwall mine output accounted for 40 percent of all underground output in the U.S., and today longwall mines account for approximately 50 percent of coal produced underground in the United States. A 53 percent increase in average shift production rates has occurred over the last fifteen years. This increased longwall productivity has meant that far more dust is being produced and controlling respirable coal dust provides an ongoing challenge for coal mine operators. The National Institute for Occupational Safety and Health (NIOSH) has completed a series of benchmark surveys at longwall operations across the country to identify current operating practices and the types of controls being used. Gravimetric and instantaneous dust sampling was completed to quantify dust generation from major sources on the longwall section and to determine the relative effectiveness of the different control technologies in use today. Substantial reductions in dust levels were realized at sampling locations on the face when compared longwall surveys conduct in the 1990?s. This paper summarizes the results from the underground dust surveys and discusses current longwall dust control technology and operating practices.

"Building and operating tunnels has been a highstakes endeavor in terms of costs, complexity, safety and impact on contractors, owners and the public for a long time. There are many variables that need to be closely monitored and managed to construct a tunnel on-time, on-budget and safely. Once built, there are also many elements that need to be monitored and managed to operate a tunnel safely and efficiently.Fundamentally, the risks to the safe and efficient construction and operation of a tunnel are a function of the degree of harm and the likelihood of the harm occurring. From a cyber security perspective, risk assessments used to inform decision makers and support risk responses are typically based on identification of relevant threats; internal and external vulnerabilities; the impact that may occur given the potential for threats to exploit vulnerabilities and the likelihood that harm will occur. This article focuses on creating awareness of vulnerabilities associated with technology used in tunnel construction and operation and some of the challenges associated with remediation of these vulnerabilities. As we all know, technology has been evolving at an accelerating pace and infiltrating every aspect of our lives in new and often exciting ways. In the world of tunneling, sensors and systems that provide position and time have become accurate, rugged and cost-effective enough to come out of the research labs and into the equipment and systems used every day. There are many opportunities to employ connected computing devices and sensors to costeffectively capture, integrate and analyze key data with minimal latency to manage risk better, improve safety and avoid surprises."

So much so that, certainly, no .discussion on metallurgy can be complete without including some reference to it.Obviously, one paper cannot hope to cover in detail the relationship of the whole science of metallurgy to the mining industry. The large massive sulphide deposits present a formidable challenge to the economic geologist. They represent such huge concentrations of iron, sulphur, base metals and precious metals that it is difficult to conceive how they were derived from the crust or mantle of the Earth. The problem of the origin of massive sulphide deposits has generated a geological controversy which has lasted two hundred years and shows no sign of abating. The economic incentive to solve this problem is great, as the massive sulphide bodies are the moat important source of the world's zinc, lead, and silver, as well as yielding large amounts of copper and gold. Using the criteria of abundance, size, age distribution and gross value of contained metals, the predominant sub-group of the massive sulphide deposits is that which comprises the crudely conformable pyritic (or pyrrhotitic) deposits which occur in stratified volcanic and sedimentary rocks or their metamorphosed equivalents. Examples of this sub-group are: the Buchans, Bathurst, Noranda, Kidd Creek, Flin Plon and Sullivan deposits of Canada; the Mt. Isa and Broken Hill orebodies of Australia; the Cyprus, Rio Tinto, Meggen and Rammelsberg deposits of Europe; Ducktown and the United Verde in United States; and the Kuroko deposits of Japan.

THE death of Sir Lowthian Bell removes almost the last of the group of heroic leaders who made their age and ours the Age of Steel-a group which his luster and the luster of his peers, Bessemer, Siemens and Tunner, made one of the most brilliant, perhaps even the most brilliant, in the annals of industry. Born on February 15th, 1816, in Newcastle-on-Tyne, he was the eldest son of Thomas Bell, an alderman of Newcastle, and of his wife Catherine Lowthian of Newbiggin, near Carlisle, in Cumberland, and was christened Isaac Lowthian, the name of his maternal grandfather. His father was born about 1788, and came in 1808 from Newbiggin to Newcastle, where he entered the office of the Cumbrian firm of Losh and Company, manufacturers of alkali and wrought-iron, at the Walker Iron and Alkali Works, at Walker, Dear Newcastle. Young Isaac Lowthian Bell was educated at Dr. Bruce's school in Newcastle, whence he went to Germany and Denmark, learning there German and Danish. On his return he studied at Edinburgh University, and then at the Sorboune in Paris. As a student he already showed the remarkable character and application which later gained him such distinction. After studying the manufacture of alkali near Marseilles, he returned, and when about twenty years old (A.D. 1836) was employed in his father's firm, then called Losh, Wilson, and Bell. After spending a year in their Newcastle offices, and after visiting the important iron-making districts of France, Germany, Poland, and Austria, he took charge of their rolling-mills at Walker, and profited so well by his opportunities, and especially by the teachings of his father and of Mr. John Vaughan (later one of the founders of the great firm of Bolckow, Vaughan & Co.), that he was made manager of the Walker works in or about

"Société le Nickel exploits the Dôme lateritic nickel orebody at its Tiébaghi operations in New Caledonia. The site geology has an obvious bearing on recovery and metallurgical performance, with controls that are extremely complex at all scales, including lithology, oxydo-reduction conditions, mineralogy and multivariate geochemistry. In this context, establishing an adapted recoverable resource estimation method that is efficient and transparent proves an interesting challenge. The method must address the key notions of: support effect – the exploration data set can only warrant the estimation of large panels of 20 × 20 × 3 m3 that are much larger than the selective mining units (SMUs) of 5 × 5 × 3 m3 information effect – the selection at the production stage uses estimates based on information that is much denser than that available for mine planning.The problem is rendered more complex in this type of deposit by the fact that the additive variables used for the estimation (metal accumulation and ore tonnage) are not the ones used to establish the selection at the mining stage (ie the nickel grade of SMUs, which is the ratio of accumulation on tonnage). Eventually, the selection criteria not only involve Ni grades, but also other elements such as Al2O3, Fe2O3, MgO and SiO2.The solution presented in this paper is to construct an SMU multivariate simulation in the saprolitic horizon and construct an efficient post-processing aimed at producing multivariate recoverable resource estimates. The efficiency constraints imposed by the number of blocks to be simulated (1.5 million) lead to resorting to direct block simulations. The underlying multivariate discrete Gaussian model, which had its validity tested beforehand, is put to good use to mimic the selection process that happens at the mining stage by offering the ability to simulate a composite value located at random within each block.The paper also presents the application of a scenario reduction algorithm to pick a representative subset of a few simulations to help appraise the risk attached to the downstream (reserve optimisation and mine sequencing) phases of the project. The original version of the paper (see Deraisme, Bertoli and Epinoux, 2014) used an early version of the scenario reduction plug-in (named S2RM) built in Isatis® software, where measuring the distance between the initial set of scenarios and the reduced set was based on Ni only. The implementation presented here is for the second generation of S2RM, where measuring the distance between the initial and the reduced set of realisations is based on all study variables. Blast exclusion zones are instrumental in protecting equipment and personnel. When there is no constraint in footprint, the exclusion zone can be enormous, practically eliminating the hazard of flyrock. Unfortunately, this may not always be the case and in many circumstances the blast design needs to be altered to suite such scenarios. When a conservative design approach is taken, it could completely eliminate any form of ejection, whether it is from the free face or the bench itself. The degree to which typical flyrock models describe fragment motion through air with ballistic flight equations are not justified when such a high level of control is applied. Atomaer (Pty) Ltd is a technology-driven company with interests in Western Australian nickel. Atomaer (RSA) Pty Ltd is a process technology company operating in Southern Africa. It is a subsidiary of Atomaer Holdings Pty Limited (Atomaer), which has operating companies in Australia, Africa, South America, and Europe. In addition, Atomaer has substantial shareholdings in Braemore Resources Plc, a nickel company with interests in sulphide nickel projects in Western Australia and Southern Africa listed on the Alternative Investment Market of London Stock Exchange (AIM); Mintails Limited, a gold company with interests in gold operations in South Africa listed on the Australia Stock Exchange; and Brinkley Mining Plc, a uranium company with interests in Southern Africa listed on the AIM. Independence Platinum Limited (IPt) has been formed by Atomaer to undertake the commercial development and exploitation of Mintek's ConRoast technology with the strategic objective of providing independent smelting and refining facilities with lower capital and operating costs for the treatment and processing of platinum concentrates, including those with high chrome and/or nickel and/or low platinum group metal values, for the development of platinum projects by junior resource companies in South Africa. The platinum mines of South Africa have traditionally exploited the Merensky Reef of the Bushveld Complex for platinum production. However, the mining of this reef is reaching depths that require significant vertical shaft systems and costly refrigeration for access to the ore bodies at deeper levels. In the past decade, all of the platinum producers have begun to mine the UG2 reef because of its cheaper mining costs. UG2 ore usually has lower platinum group metal contents and higher chrome values. The new junior resource companies have gained access primarily to UG2 reefs. Consequently, platinum mining is becoming more UG2-based and the resulting concentrates contain high levels of chromitite. UG2 concentrates are difficult to smelt in the current six-in-line submerged arc furnaces, and this leads to increased capital and operating costs for those smelting operations. The platinum mines of South Africa produce the majority of the world's platinum metal used in automotive catalysts and jewellery. As the platinum market is forecast to grow at a healthy rate into the future, the SA producers Anglo Platinum, Impala, Lonmin, and Northam are planning to expand mining operations in the next decade. Furthermore, a number of smaller new producers who are accessing other mineral resources are entering the buoyant market and will produce platinum concentrates for purchase or toll treatment. Hence, a shortage in both smelting and refining capacities is anticipated. Further smelting and refining facilities independent of the major platinum producers will be required in South Africa by 2010. Mintek's ConRoast technology is best suited to fill the need to treat a high proportion of UG2 concentrates in a cost-effective and technically efficient manner. Atomaer base metals technology has been developed for rapid leaching of nickel and other base metals from alloy. Significant reductions in capital and operating costs are expected from the combination of the two advances in process technology compared with that installed at the existing smelters and refineries that were established over 20 years ago. Independence Platinum will be listed on AIM and the Johannesburg Stock Exchange (JSE) in 2007, and developed as an independent, vertically-integrated platinum company using its independent smelting and base metals refining facilities as a strategic platform to promote the development and mining of platinum projects by junior resource companies in South Africa through technical co-operation, strategic alliances, joint ventures, off-take, toll-refining, and marketing agreements.

Climate change is the central challenge facing the mining industry, wherein producers must simultaneously ramp up production to provide the raw materials for clean technologies while transitioning to carbon neutrality. While transitioning to renewables will take time, mines can start improving energy efficiency while improving productivity today. This paper proposes a novel technology ecosystem that can help. By monitoring mine shovels, loaders, haul truck paths, and conveyor belts, the proposed technology enables particle size measurements throughout the operation, truck and belt volume monitoring, ground engaging tools monitoring, and boulder detection.

The use of electrical or electronic safety systems to protect personnel, and the engineering tasks associated with the design, manufacture and maintenance of these systems are referred to using the term ‘functional safety’. In the past ten years, the use of such systems has transitioned from its traditional base, in oil and gas major hazard facilities, to being a part of most large mining and materials handling projects. In particular, functional safety is rapidly becoming a significant component of Australian iron ore projects, typically in the form of emergency stops, remote isolation, drive through interlock, robot cell entry or mobile machine anti-collision systems. While these types of systems have been in use in iron ore for a long time, functional safety standards (eg AS61508, AS62061, AS4024.1503) require more rigourous demonstration of the level of risk reduction provided by the systems.The early engineering project development phases require a number of key decisions to be made around functional safety implementation. The decisions made at this point in a project will go on to dictate the required types of risk assessment and analysis, equipment selection, sizing of substations, and training and maintenance requirements for owners. For project teams that are unfamiliar with functional safety, or inexperienced in its application, an incorrect decision or failure to understand the requirements of the chosen standard can lead to a failure to meet contract requirements, as well as major cost and schedule overruns.This paper examines the implications of functional safety on early engineering studies, focusing on the following questions: How does a designer or company determine what standard is best suited to their project? What key aspects of functional safety design need to be considered during project development? As sponge iron is much like porous, coarse sand, it is a tempting material to use in place of coarse scrap iron as a precipitant of metals, for scrap iron is relatively awkward to handle and exposes only a small amount of surface to pregnant solutions. The results of the experiments described in this bulletin are largely a record of the behavior of sponge iron as a precipitant of lead and copper under laboratory conditions comparable to those that would obtain in a commercial plant were it attempted to secure the maximum contact of surface between solution and precipitant. Physically, the study is largely one of the rates of diffusion of the metal ions in the solutions and through the metallic coatings surrounding the sponge iron particles. The practical objectives are to remove completely the valuable metals from solution and at the same time to obtain a precipitate of good grade. In order to fulfill this latter requirement, the precipitant should be almost completely consumed. These practical ends are usually secured by having the two substances involved pass each other counter-currently-that is, in such a manner as to have the strongest precipitant in contact with the most impoverished solution, and vice versa. EXPERIMENTS WITH LEAD SOLUTIONS In nearly all of the schemes involving the leaching of lead from its ores the metal has been put into solution as the chloride, and fairly concentrated liquors have been employed. The solubility of lead under such conditions is comparatively small at best; the hot pregnant liquors from which the metal is to be precipitated are then made nearly saturated with respect to lead, and only a very small amount of acid is used. Therefore it was not as necessary to learn the behavior of the sponge iron in precipitating lead from solutions of widely differing composition as to learn under what conditions, and with what mechanisms, this precipitation might be carried out most readily.

In 1983, U.S. Bureau of Mines personnel examined mines, prospects, and mineralized areas to determine the mineral resources in the Carrizo Gorge Wilderness Study Area (WSA). The 14,571-acre WSA, located 42 miles east of El Centro, CA, is underlain primarily by Mesozoic granitic rocks and pre-Cretaceous metasedimentary rocks. Six prospects were found within the WSA and two mines are adjacent to it. Mineral deposits within the WSA have had insufficient development and rock exposures for calculating resources. The WSA contains no identified mineral resources; however, further exploration may disclose gem quality minerals. Two mines adjacent to the WSA, the Pack Rat and Beebe Hole, have produced gem quality kunzite and aquamarine from pegmatite dikes. According to the owners, a significant amount of gem minerals have been mined here in the last decade. Based on past production and the characteristic minerals indicating gem-bearing pockets, additional pockets are likely. Similar pegmatite dikes occur from Mt. Tule to Carrizo Gorge and may contain gem-bearing pockets. Pegmatite dikes also occur in the WSA near Rockhouse Canyon, but contained few of the mineral associations indicative of gem-bearing pockets. Manganese occurrences north of the Beebe Hole and Packrat Mines and within the WSA contain low concentrations of manganese. Significant sand and gravel were not identified. There is no evidence of deposits of oil and gas, or geothermal energy.

The Mississippi embayment ball clay region, including western Kentucky, western Tennessee, and northwestern Mississippi, accounted for more than 95% of the total U.S. ball clay production during 1985. Other producing states during the year were Texas and Maryland. The U.S. Bureau of Mines reported total domestic production for 1985 of 910,000 short tons. The United States exports approximately 20% of the annual ball clay production. Geologically, the ball clay deposits are sedimentary in origin and occupy the north- eastern flank of the Mississippi embayment. The commercial deposits have been confined to the Claiborne and Wilcox formations within mid to lower Eocene and upper Paleocene age strata. The deposits are lenticular in nature and highly variable in areal extent and thickness. They exhibit very abrupt lateral boundaries and are bounded laterally and vertically by unconsolidated sands, silts, and waste clays.

Open-type, or main beam, tunnel boring machines (TBMs) can achieve impressive rates of production. This type of TBM typically outperforms other types of TBMs currently in use around the world. In good ground conditions, main beam machines have achieved production rates of more than 1 km/month (0.6 miles/month). However, installing effective ground support in poorer rock conditions on main beam machines can be a major cause of concern. The design of main beam machines is such that installation of ground support equipment, particularly at the front of the machine in what is commonly known as the L1 support zone, has traditionally been limited to rock drills and basic working platforms. Personnel working in the L1 zone have, until recently, been protected by roof supports and extended fingers, often with restricted access to working areas.

"The Construction Group is responsible for the construction of all of Placer Dome’s major projects and contributes to the development of most major feasibility studies. This group includes an international collection of Placer Dome people who have built mines in Papua New Guinea, Australia, Chile, the United States and Canada. This paper outlines the changing role of the Construction Group from the early days of single projects to today, when several projects are in various phases from feasibility to construction. The projects’ total annual budgets have from time to time exceeded one billion dollars. IntroductionAs the competition increases globally between the major gold mining companies, the impetus continues to build mines at the lowest cost with the shortest construction schedule. Placer Dome has more than met that challenge by building mines around the world at a pace that was not contemplated 15 or 20 years ago. Since 1960, Placer Dome has built 30 mines and expansions at a cost of nearly 4.5 billion dollars.In the last six years alone, Placer Dome has built 14 mines and expansions, worth 2.6 billion dollars. The company’s success is due primarily to Experience, Innovation, People and Standards.The diversity between projects, let alone the pace, has been a challenge. Placer Dome has built mines around the world in a variety of extreme climates and distinctive cultures. The construction experience gained from these projects has allowed Placer Dome to develop an extensive knowledge and data base making it easier to “fast track” projects.Through the years Placer Dome has developed a human resource base. It now has an experienced and adaptable core team of people able to set up construction projects and build mines anywhere in the world. By developing standards and systems to support the projects they can act as soon as a new project is approved, and complete the projects quickly and effectively. The result is that Placer Dome now has the construction expertise to build its own mines, giving them the competitive advantage that allows them to design and construct a mine in the least time at the lowest capital cost. This was demonstrated in March 1997 with the completion of the construction of two new mines in 12 months."

The results of development of technologies for flotation concentration of two types of complex beryllium ores are presented: beryllium-fluorite in apocarbonate metasomatites (Type I) and rare-earthberyllium in feldspar metasomatites (Type II). The researched samples of Type I ores contain a mineral complex that is difficult to separate by fatty acid flotation, including phenakite, bertrandite, fluorite, and carbonate minerals. The ores are characterised by an increased content of fluorite and carbonates. The sample of Type II ore contains such rare-earth minerals as bastnesite and orthite, and a beryllium mineral - cleiophane. The size of individualised grains of cleiophane does not exceed 0.04 mm, which significantly limits the possibility of extracting the mineral into the enrichment concentrates. For the Type I ores, a technological process for the extraction of fluorite using a phosphorus-containing reagent from the class of dialkylphenyl polyethylene glycol esters of phosphoric acid (DAPEGEPhA) has been developed. The use of DAPEGEPhA enables the production of a high-grade fluorite concentrate (95% CaF2). The production of commodity phenakite-bertrandite high-grade (10% BeO) flotation concentrate from the fluorite flotation tailings with using a fatty acid collector is based on binding calcium in the flotation pulp using soda, sodium hydroxide and sodium tripolyphosphate. The use of a combined collector based on fatty acids and DAPEGEPhA made it possible to produce a collective rare-earthberyllium- lithium concentrate with high REE extraction (88%) from the Type II ores. "Eicosanoids and cylokines produced by the pulmonary alveolar macrophage (PAM) are key mediators for the defensive role this cell plays in neutralizing the inhalation of particulate dusts and foreign microbes. However, these factors are also mediators of inflammation and their uncontrolled release may cause the lung injury and liorotic changes observed in individuals exposed occupationally to mineral dust. In vitro studies have suggested a link between the production of certain eicosanoids and cylokines released from the activated PAM. To this end, we have studied the release of eicosanoids (PGE2, TXB2 and LTB4) and cylokines (IL-1 and TNF) by cultured PAM, obtained from healthy human volunteers by bronchoalveolar lavage under basal culture conditions and following acute exposure to provocative agents such as the endotoxin lipopolysaccharide (LPS) and mineral dusts including silica. Studies were also carried out in the presence of specific cyclooxygenase inhibitors. The addition of LPS (10 µg/ml) to the culture medium produced a significant increase in PAM release of the eicosanoids: PGE2 (3.5 fold over baseline), TXB2 (2.8 fold over baseline) and LTB4 (4 fold over baseline). LPS also elicited a 2.5 fold elevation in IL-1 from these cells and a 9 fold increase in TNF release. The cyclooxygenase inhibitor, ibuprofen, significantly inhibited the LPS-induced rise in PGE2, TXB2 and TNF but had only a modest suppressive effect on IL-1 release. Exposure of these cells to silica dust elicited a similar augmentation in eicosanoid and cylokine release which was neutralized by prior exposure of the cells to ibuprofen. These results suggest that the enhanced production of eicosanoids and cytokines by PAM exposed to endotoxins and/or mineral dusts may be linked and that inhibition of eicosanoid release is linked to the production of TNF. IL-1 release however is not effectively neutralized by cyclooxygenase inhibition.INTRODUCTIONThe initial pulmonary cellular response to inhaled mineral dust involves the elaboration of potent inflammatory mediators from the pulmonary alveolar macrophage (1). These mediators include the eicosanoids (oxygenated metabolites of arachidonic acid) and cytokines (intercellular peptides) which are released into the alveolar space by the interaction of mineral dust with the PAM surface membrane. The eicosanoids and cytokines promote local nonvascular and vascular smooth muscle contractility changes, elicit the recruitment of additional mononuclear cells into the alveolar space, and enhance the activation, growth and differentiation of lung fibroblasts (2). In this study we sought to examine whether eicosanoid release and metabolism are linked to cytokine release when PAM are activated upon exposure to endotoxin or mineral dusts. Modulation of these pathways through inhibitory pharmaceutical agents may help attenuate the Injurious effects caused by a sustained release of these proinftammatory intercellular factors (3). To that end, we have studied the in vitro effects of endotoxin and silica on eicosanoid and cytokine release from cultured PAM obtained from healthy human volunteers."
