Introduction Autogenous, semi-autogenous, and conventional grinding circuits are used to process ores. Current emphasis, though, is towards the use of the first two systems. The reason is the generally lower initial investment required for an autogenous or semi-autogenous circuit as compared to a conventional comminution circuit. However, the effect of operating costs that will significantly affect overall comminution economics is not considered in this evaluation. About 70% of the energy in a mineral processing plant is used in the comminution circuit. Until the 1974 oil embargo, energy was relatively inexpensive. Since that time, the cost of energy has escalated 40-fold. As a result, energy cost now forms a significant portion of operating costs in comminution. According to a survey conducted by General Electric Co. (Powers, 1982), the price of electricity will continue to rise 15% to 20% per year, in spite of the current oil glut. Both autogenous and semi-autogenous grinding circuits consume more energy than the conventional comminution circuit. Higher consumption ranging from 16% to 35% has been reported in the literature (Bassarear, 1982; Gjerdrum. 1979; Betts, 1982; and Christie, 1979). Consequently, even a small increase in energy costs is likely to have a significant impact on operating costs. Therefore, the economic decision of selecting an optimal comminution circuit is important. More than ever before, it is now critical to include in the evaluation of communition circuits not only the capital costs, but also the operating costs; in particular, the expected energy cost over the life of the operation. Failure to do this may lead to a less than optimal comminution circuit. Design A comminution system should be designed only after bench-scale or pilot plant testing of a representative sample. Conventional crushing and grinding circuits can be designed on the basis of the Bond Work Index (Rowland and Kjos, 1980). Over the years, sizing of full-scale rod and ball mills from Bond Work Index data has become well established. Autogenous and semi-autogenous mills cannot be sized based on bench-scale tests. Pilot plant tests are necessary to determine the amenability of the ore, critical size buildup, and power requirements. For this study, conventional and semi-autogenous grinding circuits were designed for ore throughput of 12, 24, 36, 48, 72, and 90 kt/d (13,200, 26,400, 39,600, 52,000, 79,200, and 99,000 stpd). Rod and ball mills were scaled up on the basis of the Bond Work Index. A two-stage semi-autogenous grinding circuit was designed on the basis of pilot plant test results. THrough the courtesy of Dr. James Douglas I am permitted to give a summary of some of the results obtained in leaching slimes tailings in a series of experiments conducted under my direction at Douglas, Ariz., during the past year. While the test run for which the data are given deals with a particular case, much of the work is general in its application and may therefore be of interest. This work was done upon the ores of the Burro Mountain Copper CO. A description of the geology of the district will be found in the paper by R. E. Somers published in this volume, p. 604. The mineral is pyrite and chalcocite very finely disseminated throughout the gangue and a characteristic of all the ores is the large amount of alumina present. A representative analysis of the particular ore under consideration would be about as follows: Cu, per cent..................... 2.35 S, per cent...................... 3.0 Ag, ounces per ton.............. 0.05 CaO, per cent.................. 0.5 Au............................ The ore would have to be quite finely crushed to liberate the mineral for leaching; concentrating after crushing would cost but a few cents a ton; and a higher total reeovery would result from leaching tailings rather than ore. 2. Rough concentration would yield a suitable material for roasting to make sulphur dioxide or sulphuric acid in connection with the leaching scheme. 3. Such a program put the question of cost vs. recovery on a basis where flotation and discarding of tailings were directly comparable. This case study is an example of how Encyle/Texas, Inc. (a subsidiary of ASARCO) responded to various socio-economic forces by converting an idle zinc hydrometallurgical refinery into a metals recycling facility. The facility accepts metal-laden process waste streams and produces various "synthetic ores" which are sold back to industry. Acceptance and reactions by industrial, political, economical and regulatory interests towards the project are also discussed.

This paper supplements and brings up to date a report prepared earlier this year and presented at the Eleventh Pan American Railway Congress in Mexico City. Most of the original report is contained in this paper in order to acquaint the reader with the project background. On December 1, 1961, construction of this locomotive was completed. Stationary testing started immediately, and a host of problems in various areas were uncovered. Many of these had no relation to the use of coal in a direct fired gas turbine. All of them emphasized the problems which plagued previous projects of this kind. In 1944 the Locomotive Development Committee of Bituminous Coal Research, Inc., was formed to study the problems of combustion, coal handling, and fly ash removal which would have to be solved before coal could be used to fire a gas turbine. By 1947 this committee was composed of the chief executive officers of nine eastern railroads and five eastern coal companies. All of the members of this committee were vitally concerned in a coal fired gas turbine because coal cost only one-fourth as much per heat unit as fuel oil. A number of research projects were started to develop and test components on a small scale. This included work at John Hopkins University, Battelle Memorial Institute, Institute of Gas Technology at Chicago, Purdue University, and Southern Research Institute. By 1946 a pilot plant had been installed at the Dunkirk, New York plant of Alco Products Div. of American Locomotive Co. The U. S. Bureau of Mines made available to the Locomotive Development Committee (hereafter called LDC) a Houdry process gas turbine plant, originally destined for a Russian oil refinery. The full scale pilot plant was used to evaluate the results obtained from the small scale studies under actual gas turbine conditions. Operation of this plant disclosed a number of problems which had not been discovered in the small scale tests. By 1951 four 250 hr tests had been conducted with the Houdry gas turbine to test out a variety of equipment. The major problem concerned the removal of abrasive fly ash from the products of combustion so that wear on turbine blades would not be excessive. The first 250 hr test showed that the original fly ash separating equipment was inadequate. Large ash particles passing through the turbine blading were nicking the leading edges of the first row blades. At the conclusion of the second 250 hr test, erosion was so severe that the first row of stator and the first row of rotor blades were replaced. At the end of the third 250 hr test, it was necessary to replace the first three rows of stator blading. The fourth test was used to test out a new design fly ash separator developed by LDC and named the Dunlab tube. This device gave more encouraging results than any of the others tested. Early enthusiasm for overcoming major difficulties had resulted in placing an order in 1946 with Allis-Chalmers Mfg. Co. for a 3750 hp regenerative gas turbine. The gas turbine, together with associated combustion and ash removal equipment, coal handling equipment, generators and gear reduction unit was installed at Dunkirk by 1951. A 750 hr test was made in 1952 with the LDC-Allis Chalmers gas turbine power plant. At the end of this test it was necessary to replace the first four rows of both rotor and stator blading. Rows 5 and 6 did not require replacement. In 1953 the American Locomotive Co. joined with LDC and assumed an active role in equipment development and manufacture. During 1954 LDC-Alco operated the rebladed gas turbine 300 hr to determine if a new fly ash separator was adequate. Performance of the improved separator was considerably better, but trouble was experienced with clogged blowdown lines from the Dunlab tubes. When these lines clogged it permitted the passage of abrasive particles. In 1955 a total of 1421 hr was run. This included 243 hr of cycle operation on Union Pacific coal from the D. O. Clark mine at Superior, Wyoming. During the 243 hr a total of 370 tons of coal was consumed. The purpose of this test was to determine if U.P. coal was suitable for use in a coal fired turbine. It was the unofficial opinion that it was as good or better than the eastern coals that had been tested.

When Caterpillar announced that it had purchased Bucyrus for $8.8 billion, it was billed as the next great chapter in the history of mining. Surely, when the history books are written, it will be remembered as a deal that changed the face of the mining equipment sector, and it could well be a major milestone in the path toward autonomous mining in the future. In the short term, the deal is a clear indication that the world?s largest equipment maker is bullish on the mining sector. The purchase of Bucyrus allows Caterpillar to expand its mining interest to six product segments (underground, surface extraction, large mining trucks, surface material handling, hauling products and growth markets) with 16 product lines to serve the mining industry. But the commitment doesn?t end there. According to Steve Wunning, president of Caterpillar?s resource industries group, which includes Cat?s global mining division, the commitment to mining goes far beyond the acquisition of Bucyrus.

"Microorganisms have the ability to remarkably adsorb and/or absorb heavy metal ions. Hence they are being studied widely as a means of removing metal ions from aqueous solutions both in order to clean up toxic streams and to recover valuable metals from these and other streams. It is often difficult to harvest organisms such as bacteria and unicellular algae since they are usually too small to be readily screened out. Froth flotation may prove to be a feasible method to harvest such organisms subsequent to the ad/absortion of the metal Ions. The present paper discusses the surface/electrokinetic properties of microorganisms and the modifier-collect conditions under which they should float. The zeta potential of the green alga Chlorella vulgaris was measured in the presence of AI(III) and Pb(II) ions and in the presence of dodecylammonium chloride and sodium dodecylsulfate. Present and past data suggest that the organisms should behave similarly to other finely divided, charged (polar) solids with their behavior sometimes modified by the non-polar surface regions present.IntroductionThe overall field of bioengineering is growing at a remarkably accelerating rate. Hydrometallurgy and mineral processing are included among those disciplines where there has been actual or proposed commercial use of bioorganisms. Further, the actual or proposed use includes bacterial enhancement of leaching of a number of different types of ores (1-5), modification of the flotation or oil agglomeration separation of minerals by bacteria (6,7) and microorganism (algae, bacteria or fungi) concentration of metal ions present in effluent or other mineral processing or hydrometallurgy aqueous streams (8-23)."

The processes involved in the concentration of magnetite ore with process water as the aqueous medium can have a strong influence on the surface properties of the magnetite particles. In this study, different leaching methods, as well as zeta potential measurements, were used to examine the major easily accessible ions on the particle surfaces and their effect on the surface charge. The results show that the enrichment of elements on the surface of the particles appears early in the refinement process. Ca was enriched up to 15 times and Si up to 50 times compared to what pore water alone would have given. The leaching experiments also showed that the addition of water glass increased the surface enrichment of Si and that the collector might hinder desorption of Ca. An alkali leaching method was evaluated, which showed that Si was much more enriched than S and Cl. Zeta potential measurements on synthetic magnetite particles showed that process water resulted in zeta potentials different from the potentials measured for these particles in aqueous calcium and silicate solutions. Therefore, it is important to consider the complexity of the process water when making predictions about the properties of the particle surfaces in the final magnetite concentrate, which most studies do not. Zeta potential measurements were also used to rule out a strong influence from organic matter in the process water on the magnetite zeta potential.

With a growing emphasis on maximizing the value by rapidly deploying it across their assets, mining firms are eager to identify any issues that may hinder such efforts and to refine their approaches to overcome them. As such a CDE vision can be considered realized when the following components are addressed and functioning as intended - People: the right resources working together and efficiently to achieve shared goals, Process: well-defined workflows mapped out for key value processes, Technology: availability of the correct tools and applications to increase efficiency in work execution and Locations: regional locations/ sites that need to be linked for collaboration and information exchange. For the above vision, an approach that considers the degree of interaction between the roles in metallurgy operations, mineral resource planning, mine engineering, plant operations, logistics and infrastructure, and which accounts for the current technology states across these groups to determine sub-processes where the impact of a CDE will be the most is critical. In effect, CDE is more about transforming an organization?s operational philosophy than a technology driven project. This paper examines the definition and components of a CDE in a mining context. Real-time operations co-ordination, maintenance interventions, production analysis and optimization processes are examined along with current state technologies and manual processes as candidates which are likely to result in most benefits from collaborative work processes. Finally, the paper examines technologies that can be used to further enable a collaborative working environment.

The Wyoming craton in the western United States is a favorable terrane for diamond deposits. The craton hosts a variety of ultramafic, ultrabasic, and ultrapotassic rocks. Additionally, a few hundred 'kimberlitic' heavy mineral anomalies and some geophysical and remote sensing anomalies indicate the presence of undiscovered, mantle-derived, pipes in the craton. The Wyoming craton is separated into a Wyoming 'archon' known as the Wyoming Province, and the Colorado 'proton' known as the Colorado Province. The 'archon' underlies much of Montana and Wyoming and continues north into Alberta and Saskatchewan; the Colorado 'proton' underlies much of Colorado and a small portion of Wyoming. Although most exploration geologists would consider the 'archon' the most favorable terrane for commercial diamond deposits, the 'proton' has produced more than 120,000 diamonds, while the 'archon' has not been very productive.

Foreword The complete history and early development of the Sullivan mine was fully covered in a paper entitled 'The Development of the Sullivan Mine and Processes for the Treatment of its Ores', prepared by the staff of the Consolidated Mining and Smelting Company of Canada, and presented at the Annual Meeting of the British Columbia Division of the Institute at Trail in October, 1924. At a subsequent meeting at Blairmore, Mr. E. G. Montgomery read a paper on the mine, and in 1926 Mr. M. M. O'Brien brought the subject up to date at the joint meeting of the American and Canadian Institutes at Cranbrook. The present paper is necessarily based largely on these earlier accounts. Discovery and Early Development The outcrop was discovered in 1892 and the property was bonded and later sold to the Sullivan Group Mining Company, of Spokane. From 1896 to 1899, surface stripping and test pits (at A) around the outcrop exposed a vien of sulphides with varying amounts of lead and zinc. In some places, a 35 per cent lead ore carrying 15 ounces of silver was found. These lenses were of commercial value. With the completion of the Kimberley branch of the Canadian Pacific railway, in 1900, some systematic development was started. A shaft l00 feet in depth was sunk (at B). Shaft-house, boiler-room, and compressor-plant were erected, and a steam diamond-drill was put into operation.

Only in recent years has any practical headway been made in the application of aerial photography to geological problems, and up to the present time its principal value to the geologist and mining engineer has been in "areal geology;" i. e., the study of formations as revealed at the surface. The object of this paper is to set forth briefly the principal advantages that may be derived from aerial photographic data in geological work and to serve in a measure as a guide to those who may be contemplating its use. Geophysical prospecting has for object the locating of mineral under the surface. Inasmuch as the geologist's chief complaint in life is a dual one—inability to see under the surface and inability to visualize comprehensively what he does see on top of the ground—geophysical prospecting coupled with aerial photography should do much to lessen his burdens. To what extent aerial photography can be made a useful adjunct to geophysical prospecting remains to be proved, but the inference drawn here is that it holds many alluring possibilities. To avoid disappointments, it should be stated at the outset that to the geologist or mining engineer an aerial photograph or mosaic is only a tool, the efficient use of which he must set out to master. Upon his skill and ingenuity in applying it to particular problems will depend largely the value of the results that he will obtain. Under the most favorable conditions an aerial mosaic, or even a few loose photographs, because of the exceptionally comprehensive oversight which they afford of field data, such as dips and strike observations at isolated outcrops, local indications of faulting, mineral and fossil finds, greatly facilitate study and correlation, save much tedious foot work, and may repay their cost many times. Even so, types of country may be found where the most skillful use of the best of pictures will produce results so meager as to render the cost of aerial photography entirely unwarranted. Between these two extremes will be found endless variations and possibilities. Hence no set rules can be promulgated. The safest procedure is to treat each case as a separate problem and to carefully weigh the relative advantages and shortcomings of aerial photography before embarking on its use.

The current Stoilensky magnetite concentrator comprises a three-stage crushing circuit followed by three stages of ball milling, grinding and concentration to produce a magnetite concentrate with 66.5% iron (Fe). The plant is built in four lines, three lines having four mills and one line with five mills, to treat 32 Mt/y of ore and produce 13.9 Mt/y of concentrate.NLMK, the owner, is Russia’s third largest steel maker and while the ore from the Stoilensky Mining and Beneficiation Plant (SGOK) provides the bulk of their supply, they still must buy ore from external sources at world prices. A decision was taken to expand the SGOK operation to supply all the NLMK concentrate, including the production of pellets.To commence the expansion, high pressure grinding rolls (HPGRs) were introduced as a fourth stage of crushing ahead of the first ball mill. This was a relatively low capital cost expansion and increased production by 12 to 15%, the cost being much less than an additional grinding line. The importance of this development was that earlier introductions of HPGRs in Russia had presented several problems so there was some reluctance to the initial concept. The even greater significance was that nearly every magnetite plant in Russia had a three-stage crushing circuit so the successful introduction of HPGRs was of interest to many operators.The design was developed by a team, consisting of Worley Parson, Mineralconsult and PiterGORproject (PGP), for the first four units to fit inside existing building structures. The first two units were commissioned in March 2016 and the next two units in May 2016, and production improved by over 12%. Introduction Research concerning all phases of hoisting practice, but more particularly the causes and prevention of hoisting rope deterioration and the improvement of safety dogs 158 for shaft conveyances, has been carried on intensively for the past three years in the Province of Ontario. Prior to the hoisting accident at the Paymaster mine on February 2nd, 1945, there had been no fatalities in the Province due to a hoisting rope failure. It was probably on this account that little original research on hoisting problems had been undertaken here and that the mining industry had been content to look to British, South African, and United States investigators for new developments in this field. Except for the Paymaster accident this might still be the case since, in any industry, it is apparently necessary for a serious accident to occur before any extensive programme of preventive research is launched. When this accident occurred, the Government of Ontario, with the close co-operation of the Paymaster officials and the mining industry as a whole, took immediate steps to determine the cause. A five-man, non-industry committee, selected from the Faculty of the University of Toronto with Dean C. R. Young as Chairman, was appointed by the Government, and a three-man Committee consisting of R. D. Parker, General Superintendent of the International Nickel Company of Canada, Limited, R. L. Healy, General Manager of Wright Hargreaves Mines, Limited, and R. E. Dye, General Manager of Dome Mines, Limited, with the latter acting as Chairman, was appointed by the Ontario Mining Association.

This paper relates to the foundation of a steel structure located at the San Martin de la Vega (Madrid) amusement park. The structure has more than 100 supports with various loading conditions, including important horizontal and uplift forces. Additionally, the settlement tolerances are very strict. A massive gypsum formation with interbedded clay layers and random solution great voids at various levels constitute the soil. The foundation design has been conditioned large enough these difficulties (load conditions and soil problems). The piling machine used was powerful to drill through the soft rock (UCS up to 11Mpa). The integrity of each pile had to be guaranteed by continuous recording of drilling and concrete pouring parameters. In order to fulfil all those conditions, a STARSOL type-piling rig was used. More than 4500 meters of piles were performed. The diameters of the piles were 450 and 550-mm and their depths varies from 6 to 23 meters. All piles were reinforced over their full length.

For the purpose of examining a specific problem, it is important that we look at the context, to obtain an appreciation of the general area of which the particular problem forms a part. It is my task today to speak on a particular relationship between the nations of Canada and the United States of America. What is the background? Almost two hundred years ago we both belonged to a family of nations -- the British Empire. There was a serious family quarrel in 1776 and the United States left the family and set out to make its own independent way in the world. Canada, which was not as grown up as the united States, remained with the mother country, and in 1812 resisted a vigorous attempt by the United States to get it to leave home too. With the passing of the years the bitterness of that family quarrel has faded and, aside from occasional differences, we have been getting along very well ever since.

"In the context of today's continuously changing world, there are many factors which must be considered when attempting to assess the ability of Canada's primary copper producers to remain competitive in the face of increasing competition in world markets. In this paper, we have attempted to document those factors that will have the greatest impact on the future viability of Canada's copper industry, and to suggest possible strategies to fully utilize whatever inherent advantages Canada may have to enhance its competitive position. Technological factors related to the efficiency of precious and by-product metals recovery, energy type and utilization efficiency, production and sale of sulphur by-products, and control of 502 emissions are given high priority in this evaluation of Canada's future needs to remain competitive. Particular emphasis is placed on assessing the relative viability of hydrometallurgical versus pyrometallurgical processes for copper; to address uniquely Canadian conditions and problems. Due consideration is also given to the economic incentives required to allow Canada's copper producers to compete under anticipated world market conditions and to facilitate the continued development of Canada's copper resources. IntroductionWhen we try to assess the viability or competitiveness of any industry the bottom line is still usually, ""Can we produce a pound of product cheaper than the competition?"" Canada's copper industry is certainly no exception. However, as with most resource-based industries dependent upon exports, there are many factors which have a significant effect on the cost components which make up the total cost of producing and selling a pound of copper. It is desirable therefore to have as clear a picture as possible of the cost components, the major factors affecting them, and how they compare with the competition, particularly the third-world copper producers.In this paper we have attempted to provide a brief outlook for world copper markets and a brief overview of Canada's competitive position. We have also attempted to document those factors which will have the greatest impact on future production costs, and have suggested possible initiatives to utilize inherent Canadian advantages to enhance our competitive position. The following description of the Alabama coal-,fields is taken (with slight omissions) from the report " On the Warrior CoalField, by Henry McCalley, A.M., C. and M. E., Assistant State Geologist:" The coal lands of Alabama are of the great coal basin of the Ohio, or of the great Appalachian coal-field, and are estimated at 8660 square miles. They formed one connected field prior to the great Appalachian revolution, when there was pushed up through them long, narrow, anticlinal ridges which have been denuded into the present well-known anticlinal valleys, and thus this great field has been divided up into three more or less distinct parts. These parts were named by Prof. Tuomey, in 1849, the Warrior, the Cahaba and the Coosa coal-fields, respectively, from the names of the rivers which drain them. They are of very unequal size, the Warrior, with an area of 7810 square miles, being nearly ten times as large as the other two combined, and about two-thirds as large as the coal-area of Great Britain. This Warrior field is the most northwestern of the three coal-fields of Alabama, and comprises all of the coal-measnres in Alabama, northwest of the Alabama Great Southern railroad, or all of those drained by the Warrior and Tennessee rivers. The greater part of this field, almost all of it away from the present railroad lines, has never received more than a surface-examination. Still, we have in the Warrior coal-field one of the richest coal-basins of this or any other country. As a whole, the Warrior coal-field is a broad, shallow, tray-shaped depression, sloping towards the southwest, and with its southwest end covered by a newer formation and its southeast side, especially, complicated by folds and fractures. As is the case with all of our Coal Measures. it is composed of a series of sandstones. conglomerates, shales, slates and coal-seams, with some little limestone at several horizontal positions, but contains no thick bed of this last rock like the Measures of Pennsylvania and other States. THERE are three active mines in the Chewelah-Colville district, the United Silver Copper near Chewelah, and the Old Dominion and the Chloride Queen near Colville. The Admiral near Valley is also doing development work. The Chewelah district is in the south central part of Stevens County in the Colville Valley. Chewelah, a thriving town of 1600 people, and the scene of operations of the Northwest Magnesite Co., is about the center of the district. It is situated in a farming community on the Spokane-Nelson branch of the Great Northern railway, about 65 miles north of Spokane. Only two or three of the mines in the district have been extensively developed, although many have been prospected. The general elevation of the valley, is about 1600 or 1700 ft., that of the mountain ranges on each side about 3000 to 4000 ft., with gentle slopes and rounded glaciated tops. Prospecting is often difficult because of much glacial debris.

A large open pit iron ore mine in Brazil, which has a heterogeneous lithology, faces a challenge in defining drill and blast patterns to achieve the expected results. The mine needed to obtain an automated representation of the fragmentation curves and correlate them to the respective blasts, lithologies and quality parameters, seeking the continuous optimization of the fragmentation process and its benefits to the subsequent comminution processes. Trials were carried out for 5 months to demonstrate the functionality of an autonomous fragmentation monitoring system that analyzes the granulometric curve of the material throughout the muckpile and makes the results available online and in real time. The system, which contains a binocular camera, a processing unit and an antenna, was installed in a shovel, to evaluate the following KPI's: monitoring failures, monitoring capacity, physical availability and reportability of oversized boulders. The system analyzed 33,060 samples, with an average capacity of 285 photos every 24 hours, 127 during the day and 158 at night, all measured, analyzed, stored, and made available on its online platform, reporting 223 alerts of P80 above 400mm (15.7 in) and 38 particle sizes above 900mm (35.4 in). The inactive and operational hours of the monitoring system corresponded, respectively, to the idle and worked hours of the shovel, according to dispatch data. There were no camera or processor failures, and rain, dust or poor internet conditions did not impact sampling and fragmentation analysis, indicating 100% physical availability of the system. The autonomous monitoring analyzed 139 times more samples compared to the currently used manual method. In addition to increasing sampling representativeness and automating the process, it increases the safety, quality, and productivity of granulometric analysis, eliminating the exposure of people in the mine and human bias in the analyses. Additional analyses were performed by correlating digitally centralized drill and blast data, data from autonomous drills and fragmentation results. Automated obtention of granulometric curves favors quick decision-making and optimization of blasts to achieve better results in productivity and cost reduction in the plant and mine operation. The autonomous fragmentation monitoring system met the need presented by the mine, proving to be a key tool in continuous improvement projects, including Mine to Plant.

"The inhalation of silica has been shown to produce a dramatic inflammatory and toxic response within the lungs of humans and laboratory animals. A variety of cellular and biochemical parameters are used to assess the silica-induced lung injury. The purpose of this paper is to introduce the use of luminok:Jependent chemiluminescence as a new method to study inflammation in both phagocytic cells and lung tissue recovered from silicaexposed animals. Chemiluminescence, or the emission of light, accompanies the release of reactive forms of oxygen when phagocytic cells are challenged. In this study, male Fischer 344 rats were intratracheally instilled with either silica (10 mg/100 g bw) or saline vehicle. One day after the instillations. a marked increase in the chemiluminescence was observed in the lung tissue and bronchoalveolar lavage cells recovered from the silica- treated animals when compared with the saline controls. The light reaction was markedly decreased by either superoxide dismutase or N-nitro-Larginine methyl ester hydrochloride. Superoxide dismutase is involved in the enzymatic breakdown of superoxide anion. while N-nitro-L-arginine methyl ester hydrocholoride, a nitric oxide synthase inhibitor, prevents the formation of nitric oxide. When superoxide anion and nitric oxide react. they form the highly oxidizing substance peroxynitrite. This study then implicates peroxynitrite as an agent that may be responsible for some of the oxidant lung injury that is associated with silica exposure. The use of luminol-dependent chemiluminescence may prove valuable as a method to measure the earliest events in the inflammatory process. and may be an adjunct in studying the mechanisms that produce inflammation. - Environ Health Perspect 102(Suppl 10):37-42 (1994) IntroductionResearch on inflammation has not produced a method that links the initial events of the process to the controlling events. Rather, a variety of parameters are measured that are the result of and not the basis of the inflammatory reaction. Parameters such as tissue weight, edema, blood flow, enzyme and protein release, or production of metabolites (prostaglandins, leukotrienes) have been used extensively. A method is needed that could be applied to cells of a particular type, groups of cells of different types, and even tissue, in an attempt to study the kinetics of the inflammation as well as being sensitive enough to monitor the early events of the process . Since inflammation is so complicated, the relationship between the control of the initial events and the oxidation process has never been explained nor studied in any derail."
