"SJVN Limited successfully commissioned its Rampur HEP (412 MW) in 2014. The project has been planned to operate in tandem by directly utilizing the Tail Water of SJVN’s prestigious Nathpa Jhakri Hydro Power Station 1500 MW located upstream. The initial filling of voluminous water conductor system of RHEP with 10.50 m diameter, 15 Kilometer long Head Race Tunnel, involved more than 1.5 million m3 of water. The filling operation of such a large diameter tunnel constructed in young folded Himalayas which always offer less ideal sites for tunnelling and their subsequent operations under pressurized conditions is worth sharing with tunnelling fraternity. The paper covers meticulous planning, execution, precautions taken, and performance monitoring during filling process keeping in view the fragile geology, low rock cover zones, shear zones in close vicinity of thickly inhabited areas, squeezing zones, hot water zones encountered during excavation.1.0 RAMPUR HYDRO POWER STATION IN BRIEFRampur Hydroelectric Project (412 MW), is located in Himachal Pradesh (India). This project is a downstream development of Nathpa Jhakri Hydro Power Station (1500 MW) constructed on river Satluj and commissioned during 2003. Both the projects today are now successfully working in tandem. The upstream NJHPS has been constructed on the left bank of the river while the downstream Rampur HPS is located on right bank of the river (Figure-1).The major length of HRT of Rampur project lies on right bank of river before it crosses under the river in its initial reach (Figure-2). The main features of the project are as below:• 10.50 m diameter 15 km long HRT ending in a 155.75 m deep, 38.00 m diameter surge shaft.• 3 steel lined pressure shafts, 5.4 m dia, 211-220m long.• Surface valve house 69 m long x 10.50 m wide x 23 m high housing 3 No. butterfly valves• 3 No. surface penstocks 5.4m dia 368-382m long, bifurcating into six penstocks, each of 3.80 mdia. to feed six generating units of surface power house 158 m long x 24.50 m wide x 48 m high, with Francis turbines each of 68.67 MW capacity."

The Bureau of Mines heated at 150° to 350° C for 0.5 to 360. days Green River oil shale that had been previously extracted by benzene to provide information on in situ processing conditions, to determine the effect of low-temperature heating on the unconverted kerogen, and to determine the composition of benzene-soluble thermal products. The methods used in determining the composition of the soluble thermal products consisted of fractionation of the soluble material, elemental analysis of the soluble products, fractionation of the polar resins by ion-exchange chromatography, gas chromatographic analysis of [n]-alkanes3 and mass spectral analysis of the branched plus cyclic alkanes and aromatic oils. The methods used to determine the effect of low-temperature heating on the unconverted kerogen remaining in the oil shale heated residues consisted of carbon-hydrogen analysis, volatile material determination, carbon residue determination, and oxidation rate determination. In general, it was concluded that significant increases in the degradation of kerogen to oil occurred between the temperature range of 250° to 350° C; that the composition of the soluble products depends upon the temperature at which they are formed; and that the [n]-alkanes are pyrolytically degraded from the kerogen structure. In particular, the 250° C heating temperature is beneficial to the formation of branched plus cyclic alkanes. Also, the heated and unconverted oil shale kerogen and the soluble thermal products become more aromatic as the temperature increases from 150° to 350° C.

Gangue minerals can often have severe adverse effects on selective value mineral separation. Such effects can be mediated through a variety of pathways or mechanisms. One popular mechanism is that of ?slime coating? or heterocoagulation, wherein the gangue mineral fines are believed to coat the value mineral surfaces and thereby reduce selectivity and floatability. Another pathway that has received relatively little attention is that mediated through the effect of gangue minerals on pulp rheology, and consequently selective separation. Our recent work in developing a scientific understanding of these effects has highlighted the prevalence and importance of this pathway. In this study, the impact of shape and morphology of serpentine gangue minerals on suspension rheology and selective Ni flotation from ultramafic ores was investigated. We found strong evidence for micro- and macro-network formation in the presence of acicular (needle-shaped) or fibrous minerals, which impacted pulp rheological properties. Results also supported the hypothesis that morphology has a greater role to play than mere shape. The size of the network ranged from 5 microns to 2 cm depending upon the amount and aspect ratio of fibrous minerals. These networks impede many flotation sub-processes, such as gas dispersion and bubble-particle interactions. Additionally, the small pore size of the networks prevents bubble penetration leading to a large transport of the gangue minerals to the concentrate via lifting of the entire network by bubble flux, which resulted in a significant grade dilution. Based on the scientific insights generated in this study various techniques have been developed to improve selective value mineral separation. Excavation and shoring adjacent to the above-grade Gilmore train station and elevated guideway was undertaken in 2019. A perimeter secant pile cut-off wall was constructed to a depth 37 m (121 ft) below ground surface consisting of 1.0 m (3.3 ft) diameter drilled piles spaced 750 mm (2.5 ft) on-center, tied back with soil anchors, and reinforced with W610 steel sections every 4th pile. A bracing/jacking system was also constructed to support 3 free-standing guideway caissons within the excavation. An automated instrumentation program was installed to monitor movements of shoring walls and train infrastructure in real time and consisted of a robotic total station, inclinometers, tiltmeters, strain gauges, extensometers, and anchor load cells. Baseline monitoring was able to establish diurnal and seasonal movements within the structure, strongly correlated with temperature. The system actively monitored various shoring elements and train infrastructure in real time and alerted the project team when values exceeded predetermined threshold values. Remote reconfiguration of the automated system allowed for more frequent monitoring during jacking of Pier 29 in March 2022. Monitoring data was in agreeance with on-site manual surveying results and jacking dial gauges, and proved to be a reliable monitoring system for construction applications.

The tragic incidents at Sago and Darby coal mines in 2006 pointed out the need for stronger seals to resist the blast effects of violent gob explosions. Mine Safety and Health Administration (MSHA) has published the final rule on Sealing of Abandoned Areas in April 2008. According to the final rule, the mine seals for unmonitored gob areas should withstand an instantaneous pressure of 120 psi. Reinforced concrete (RC) seal is one of the feasible options that can be used to meet the final rule. Concrete is a heterogeneous, cohesive-frictional material and exhibits complex non-linear inelastic behavior under multi-axial state of stress. The increased use of concrete as primary structural material requires the development of sophisticated material models to have an accurate prediction of the material response to a variety of loading situations. The constitutive modeling of concrete has advanced considerably in the past three decades. Choosing a reasonable concrete material model is important in order to obtain an accurate mine seal design. This paper attempts to address the effect of different concrete material models on the response of RC seal subjected to dynamic loads from methane explosion. Three concrete models, from ABAQUS library, were selected to address the effect of non-linear behavior of concrete and damages due to tensile and compressive stresses. In addition, a typical and verified concrete material model was used for conducting 3D finite element models simulating the RC seal by ABAQUS, EXPLICIT ver. 6.6.

Biological leaching using native heterotrophic bacteria was studied by the U.S. Bureau of Mines as a means of extracting manganese from a domestic low-grade wad ore. Column heap leaching simulation techniques were employed using molasses as the nutrient source for the bacteria. These column studies were designed to investigate the effects of molasses medium flow rate, concentration, and replacement interval on the extent and rate of manganese extraction. .Results showed that flow rates between 0.41 and 8.1 L/min/m2 had little impact on manganese extraction. The total amount of molasses used did have a direct effect on the extent and rate of manganese extraction. Manganese extractions over 90-pct were obtained in 12 weeks of bioleaching with 2-week medium replacement cycles.

This report presents a representative cross section of methods success- fully used to ventilate continuous-miner places as well as various factors that should be considered when selecting auxiliary ventilating equipment. Line brattice can direct fresh air to the active face reasonably well, provided that adequate area for installation is available. Continuous mining equipment, however, often restricts clearances to such an extent that proper installation of line brattice is impossible. Consequently, in many instances face ventilation is inadequate; therefore, many methods of auxiliary ventilation for continuous-mining equipment have been developed and have been discussed in this study.

The paper describes the propensity of tin to form intermetallic compounds with microalloying elements such as Nb, Ti and aluminium, which are commonly used as additions to achieve particular combination of strength and toughness. Two series of alloys were prepared by arc melting under an argon atmosphere. Each series was based on Fe, Sn and A1 with either Nb or Ti additions. The microstructures were studied using scanning electron microscopy, transmission electron microscopy, energy dispersive X-ray spectroscopy and microprobe analysis. It was found that the level of Nb addition has an effect on the formation of Sn-rich compounds. For small Nb additions, no Sn containing compounds were found, whereas in higher Nb alloy, a Laves phase (FezNb, hexagonal, a = 0.4830 nm and c = 0.7879 nm) containing about 1 at % Sn was found. A FezNb3 phase (cubic, a = 1.1261 nm) associated with the formation of alumina was also found but did not contain tin. No Sn containing compounds were found with Ti additions. The experimental observations have been compared with thermodynamic calculations performed using MTDATA.

NCHRP Synthesis 418 (2011) entitled, “Developing Production Pile Driving Criteria from Test Pile Data” details the occurrence of dichotomy between pile Skin resistance and pile Tip resistance mobilizations at different times for different hammer blows during pile driving operations. With increasing usage of High Strain Dynamic Testing (HSDT) the engineering community started paying more attention to this occurrence of dichotomy resulting in overdriving, pile damage, increased claims, and loss of time. To overcome the dichotomy limitations of HSDT, NCHRP recommends, “the use of superposition to combine the back-calculated tip resistance from a blow on initial drive or from redrive with the back-calculated skin resistance from the early blows at the beginning of restrike”. However, the current practice of external mounted gauges and signal matching for a single blow are unable to evaluate independently the Skin resistance and Tip resistance to achieve the goal of determining the optimum Total Pile Capacity using the superposition. This paper discusses advancements of science and technology with sensors embedded at the pile top and pile tip, the data collected is analyzed by the state-of-the-art FDOT Method to determine Skin and Tip resistances independently for every blow. Independent Tip and Skin resistances each with a Resistance Factor can be summed up to obtain optimum Total Pile Capacity. Additional benefits of determining independent Skin and Tip resistances include obtaining lost skin capacity due to scour, due to pre-drilling/preforming and uplift tension capacity. Furthermore, the benefits of embedded sensors include increased quality control/quality assurance through 100% testing, accelerated construction, life cycle monitoring and asset management.

We in Kennecott are well aware that Alaskans are interested in the progress of our project at Ruby Creek. We would very much like to be able to present you with a final report at this time and tell you about a new mine. I'm afraid, however, that it will be several years at best before such a report can be made. This, then, is an interim report to describe briefly some of our findings to date, and to acquaint you with some of the problems, both geologic and economic, which must 5e resolved favorably before we can talk about a mine. There are several things about this project which make it unusual and interesting to those of us who have been connected with it. Firstly, there is the fundamental importance to Alaska, in the present stage of its economic history, of any undertaking which offers promise of turning a natural resource into a competitive export product. I am sure all of us would get a lot more satisfaction out of having a part in bringing in a new mine in Alaska than we would out of adding another one to the long list in, say, Arizona.

Land-based nickel resources include nickel sulfide and nickel laterite. With the consumption of high grade nickel sulfide, use of nickel laterite has received more and more attention. The mineralogy and sintering behavior of limonitic nickel laterite with high iron and low nickel and silica was studied to offer technical support for producing ferronickel through sintering–blast furnace route. The mineralogy results showed that the main phases in this kind of nickel laterite are goethite (FeO(OH)), gibbsite (Al(OH)3) and NiFe2O4. The TGA (Thermogravimetric Analysis) and DSC (Differential Scanning Calorimetry) revealed hydroxide minerals in addition to absorbed water. After the sintering experiments the chemical composition, phases present (XRD analysis) and physical properties of the sinter were studied. The content of FeO in the sinter increased with increasing basicity up to 1.3, and then decreased with further increase in basicity. The yield of sinter increased (from 73% to 80%) with increase in the basicity from 1.1 to 1.7, and then the yield decreased with the further increase in basicity. Mg(Ni)(Fe,Al)2O4 is the main phase in the sinter while MgCaSiO4 and Fe2SiO4 is the main binder phase when the basicity is 1.1~1.5, and MgCaSiO4, Fe2SiO4 and SFCA is the main binder phase when the basicity is 1.7~2.1.

Most of the Iron ore washing plants set up in India in the earlier days consist of sizing of the ore by dry / wet screening, washing and classification by screw classifiers. In this classical approach, iron values were lost in the form of fines and ultrafine into the tailing ponds as they had little commercial value in those days and accumulated in huge quantities over the years. As the high grade deposits are getting exhausted and the demand for high grade finer material for pellet making is ever increasing, focus is shifting towards recovering the values from the erstwhile tailing ponds by column flotation. This is also supposed to mitigate to a certain extent the environmental problems caused by the ever expanding and unmanageable tailing dams. A case study is presented wherein a composite sample is prepared from samples drawn systematically from multi - locations in a sprawling tailing dam. Laboratory scale column flotation tests on this composite tailings sample, basically originating from two operating iron ore beneficiation plants of JSW Steel Ltd., one of the leading producers of steel in India are found to be encouraging. Desliming followed by reverse and cationic flotation tests using flotation column resulted in the concentrate of 61.88% Fe, 4.81% SiO2, 2.52% Al2O3 and 3.30% loss on ignition (LOI) from the tailings analyzing 57.86% Fe, 7.10% SiO2, 3.52% Al2O3 and 6.14% LOI with 52% weight recovery. The causes for the quality improvement could be attributed to desliming of un-liberated ultra fines of kaoline and hydrated iron oxides and their further reduction by efficient flotation process. The process and the cationic collector developed for this purpose are adopted in the new flotation plant created to treat these tailings. "Qatar’s capital Doha is undergoing major infrastructure improvements including a multiphase stormwater management program. Although located in an arid climate, Doha experiences flooding during rainfall events. Dewatering operations and rising groundwater levels add to the management needs. To address those needs, construction of the Musaimeer collection tunnel along Doha’s southern rim is being followed by a marine outfall tunnel connecting a pump station to a dispersion field 10 km offshore. Qatar’s geology is characterized by sabkha deposits, a series of predominantly weak and karstic rock strata that creates challenging conditions for mechanized tunneling, as highlighted by the uncontrolled flooding of a tunnel boring machine (TBM) used on the Doha Metro tunnels. Risk associated with these challenges is magnified for the outfall tunnel by drive length, sub-sea elevation, and a single access point. This paper describes the outfall exploration strategy and design decisions made in light of experience gained during the collection tunnel construction. CONTEXT – STORMWATER MANAGEMENT PROGRAM Qatar has an arid climate with an average annual precipitation of less than 100 mm. Precipitation is mostly confined to the winter months when rare, but occasionally heavy rainfall events easily lead to flooding in Doha’s metropolitan area. Its coastal location and flat topography a few meters above the sea level mean that the depth to the groundwater table is generally shallow. The karstic aquifers in the regional dolomite and limestone series contain groundwater of high salinity, which is not suitable as a potable water source. For supplying Doha’s population of more than 900,000 (2014) with potable water, the Ras Abu Fontas seawater desalination plant is operated about 10 km south of the city. This potable water production capacity is crucial for Doha’s development and – in conjunction with overirrigation and leaking sewage tanks – is also the root cause for rising groundwater tables that have been observed over the past years."

Compilation of data on materials of metallurgical interest is one of the important activities of the Berkeley Thermodynamics Laboratory of the Bureau of Mines. Recent renewed interest in metallurgy of vanadium renders the present compilation a timely guide for experimentation to improve present methods or to develop better methods of extractive metallurgy. Thermodynamic data for vanadium and its inorganic compounds have been critically surveyed, selected, and compiled into one convenient reference from which information can be readily obtained for evaluating heat balances, determining trends, and establishing limits of metallurgical processes. By indicating the direction of experimentation, this report enables the saving of time, effort, and expense which would otherwise be necessary for trial and error experimentation. Much of the data has been obtained experimentally at the Berkeley laboratory. Other data were collected through an extensive survey of worldwide literature through April 1965. Estimates were made when no data were avail-able. Data on the vanadium halides was sparse, and the results derived from existing data were at times divergent. Some of the selected values for the halides may need modification as more or better experimental data become available.

This paper describes measurements of current and voltage done on submerged arc furnaces. The aim is to focus on and attempt to separate from one another the electric arc current and the current bypassing the arc through the charge. Toward this goal two measurement campaigns on AC-arcs burning on coke beds are ac-counted for. The first was done on a 150 kVA pilot furnace at SINTEF in Trondheim Norway, and the second one was done on a 48 MVA industrial furnace run by Icelandic Alloys in Iceland. The measurements were compared to simulations employing an advanced MFD arc model. The main results are that a coke bed does not necessarily behave as a linear conductor. However the warmer chemically transformed part of an operating Si-metal or FeSi furnace probably exhibits linear behaviour and passes a considerable fraction of the phase current, perhaps on some operating conditions, as much as 50%.

Since both rotary and jet piercing drills are used by the Iron Ore Co. at Smallwood, it is often desirable in planning to know in which regions of the orebody or new orebodies a particular drill will be the most economic. This makes it necessary to establish a correlation between drillability and pierceability and some physical rock properties. For rotary drills a good correlation was found with penetration rate and grinding factor index. The jet piercers were found to have a reciprocal relationship in the sense that the best rotary ground was the worst jet ground and vice versa. It is also indicated how an economic comparison could be made using these penetration rate versus grinding factor index curves, the hole size distribution curves for single pass and chambered holes and the mine distribution curve for grinding factor index. A discussion is presented on the fuel oxygen ratios to be used in jet piercing and on the site gas sampling and analysis which has been used to set up the drills. The fuel has been cut back so that stoichio-metric conditions exist, carbon monoxide is drastically reduced and pop-up or exploding holes eliminated. No decrease in penetration rate has been observed contrary to the published results of previous workers. The blasting procedure and results at Smallwood are discussed and the operation of Iron Ore Co.'s slurry pump-mix truck is also described briefly. Smallwood mine is part of the Iron Ore Co.'s Carol Lake operation and is situated in Labrador, 240 miles north of Sept-Iles, Quebec. Last year 15 million tons of crude ore were crushed to yield 6.3 million tons of concentrate and pellets. This year the figures will be 17 million tons of crude and 7% million tons of concentrate and pellets which is the full plant capacity. Carol Lake ores consist primarily of specularite and magnetite mixed with quartz. For convenience the ore has been split-into the following classifications depending on the percentage of magnetics in the sample, shown in brackets: specularite (0 to 10%), specularite-magnetite (10 to 20%), magnetite- specularite (20 to 30%), magnetite (>30%). The order of classification also represents the order of increasing grinding difficulty - the specularite generally being the easiest and the magnetite the hardest. The orebody also contains a small percentage of waste materials consisting of limonite carbonate, quartz carbonate and quartz magnetite. The first two materials are among the softest in the mine, generally softer than the specularite, and the quartz magnetite is amongst the hardest. The bulk of the material in the mine is of the specularite-magnetite and magnetite-specularite classifications. As a result of test drilling at Smallwood in 1960 with rotary, jet and percussion drills, the Iron Ore Co. purchased four JPM-4 jet piercers for the bulk of production drilling and set up an oxygen plant to supply 20 tons of oxygen per day. This oxygen is sufficient for two machines operating full time and one part time. In addition, there are two 50-R, one 60-R and one 40-R machines in use. The benches are 45 ft high and 50 ft holes are generally drilled. JET DRILLING At the onset of jet drilling in the late fall of 1962, two major problems were encountered: 1) freezing due to winter operations; experience and the use of heat at more places, such as the rotary head, has eliminated this,'" and 2) exploding or "popping" drilled holes; this happened frequently (several holes "popping" each day) and was the cause of two lost time accidents. In one instance a hole was being measured with a tape which fell down the hole causing it to "pop." Safety glasses though pulverized saved the wearer's eyesight. Various methods were then employed to detonate the holes before measuring or loading (dropping lighted rags of fusees down, or sparking across a spark gap). These methods were time consuming and far from completely successful. Consideration was given to the fuel oxygen ratio on the machines and what this would produce in the way of product gases. A fuel oxygen weight ratio of 0.35 which was quite oxygen negative was being used. Theoretically appreciable carbon monoxide would be produced at this fuel oxygen ratio. On the close down procedure of the jet which calls for low oxygen after flame out, oxygen would be left in the hole along with this carbon monoxide. This is an explosive mixture. "It has been discovered that copper concentrates can be leached in strong (40-60%) sulfuric acid with nitric acid, or with oxygen if nitric acid is present as a catalyst. Chalcopyrite is rapidly decomposed at temperatures well below the boiling temperature of the lixiviant (-150 C.) and the resulting copper and ferric salts are precipitated as CuS04.H20 and FeH(S04)z.4H20. Other metal sulfides such as zinc, lead, nickel, etc. are also converted to sulfates that are nearly insoluble. Sulfur is partially converted to elemental and partially to sulfuric acid. Reduction products of nitric acid can be recovered from the distillate with good recovery. Conversion of as-received copper concentrates is largely complete in about 1 hour. Introduction9 molar sulfuric acid is an unusual hydrometallurgical lixiviant: it has a boiling point of about 150 C. and a low solubility for base metal sulfates. Below about 6 molar acid, the boiling point falls below 120 C. and yields much higher sulfate salt solubility, while above 12 or 13 molar acid and at higher temperature sulfuric acid becomes an oxidizing agent, yielding gaseous S02' Thus, sulfuric acid within this range of concentration is capable of acting well above the temperature of boiling water without requiring autoclave conditions, and if typical oxidative leaching conditions are applied to suspended mineral sulfide concentrates, it is possible to obtain a conversion of these sulfides into 'crystalline sulfates. Thereafter, the sulfates can be separated by filtration or by solid - bowl centrifuge and the solution can be recirculated to the reactor vessel. Meanwhile, the solid sulfates can be treated in a typical hydrometallurgical circuit, i.e., dissolved in a return stream that is essentially water, separated from each other by, for example, solvent extraction, and recovered as metals by electrowinning."

"The complex planning required to achieve any planned production yields multiple and dissociated responses over the life of project (LOP). The customary decision-making process negates the possibilities of developing optimized and integral solutions from operating levels up to business and strategic levels. This results in an inefficient project or overall underperformance of the operation.In the project setting, disconnected trade-offs and improved performance studies become inadequate, because they are site-specific and use only a few comparative parameters. They are often based on a small data-set.Decisions taken to achieve overall objectives require the correct determination and dissemination of key performance indicators (KPIs). The KPIs indicate efficiency in operation, whereby the marginal rate of technical substitution corresponds to the rate of production. It is the necessary condition for optimization. Performance realization for sustainability follows optimization modelling, the latter often being presented with financial results such as business net present value (NPV). It considers techno-financial constraints and provides a strategic and aligned outlook of KPI decisions for an enhanced and integrated outcome.This paper explains a mining case study that is rendered efficient, and through business optimization (BO) short-term and sustainable targets are reached. Business optimization increases the NPV by 14.7%. This is achieved by challenging conventional decision-making associated with maximizing business value by including environmental externalities within the LOP."

Hydrometallurgy has a well-known and established role in the production of manganese metal (1) and manganese compounds, including battery grade manganese dioxide (2). Also, several leaching processes have been developed and applied at various times for the recovery of manganese from low grade ores (3). Recently, interest in the hydrometallurgy of manganese has been stimulated by the problems presented in the processing of sea nodules (4). Fumes and dusts from the ferro-manganese furnaces also offer a potential field for the application of manganese leaching techniques (5). In treating low grade manganese ores a major requirement of the leaching process is that it should dissolve manganese from a mixture of manganese and iron oxides. The most selective reagent for this purpose is ammonium carbamate applied after preliminary pyrometallurgical reduction of the ore (6). Since soluble carbamate complexes of the postulated type (7) are formed by both manganous and ferrous, but by neither manganic nor ferric, ions, selectivety of manganese leaching is achieved by control of the reduction operation to yield, essentially MnO and Fe304? A potential improvement to this two step process would be to simultaneously reduce and selectively leach the manganese in one hydrometallurgical operation. This possibility has been experimentally studied and the results are described below. "A sensitive colorimetric assay for detecting mucins and glycoproteins rich in 0-glycosidically linked saccharides is reported. The method combines the susceptibility of N-acetylgalactosamine terminating 0-glyco idically linked saccharides to P-elimination with the Morgan-Elson reaction for N-acetylhexosamines with free reducing ends. All mucin and mucin-type glycoproteins but none of the serum-type glycoproteins tested resulted in characteristic color production. All mucins tested gave linear responses in the range 5 to 200 ""g and the assay was also adapted to the microscale involving the use of 96-well microtiter plates. The microassay in which the volumes of samples and reagents are scaled down 2.5-fold was particularly useful in monitoring of mucins, in the presence of other glycoconjugates, in large numbers of samples obtained during fractionation procedures. Cesium chloride, cesium bromide, potassium thiocyanate, and various detergents do not interfere with the colorimetric determination. Guanidine hydrochloride, cesium triftuoroacetate, and B-mercap- . toethanol decreased color by 30 to 45%; however, the interference was not serious to prevent the use of the method for detection of mucins in their presence. The use of the method for the specific detection of mucin during fractionation by gel filtration and density gradient centrifugation of cystic fibrosis sputum samples is demonstrated. Mucins are an important class of biomacromolecules whose structure-function relationship is poorly understood (1,2). The investigations of mucins are hampered by difficulties in purifying these very large size molecules which appear to avidly bind other components of the mucus (3,4). One of the problems during purification is the specific detection of mucins. For example, during the various purification steps, the fractions containing mucins are typically detected by assaying for neutral sugars (hexoses) (5,6), for sialic acids (7), or for periodate oxidizable saccharide units (8). However, these methods are not entirely satisfactory because neutral sugars, sialic acids, and periodate-oxidizable saccharides are also components of other glycoconjugates. In addition, the neutral sugar and sialic acid measurements are also subject to interference from high levels of proteins and nucleic acids."
