Anglo Gold AshantiÆs Sunrise Dam operation is located in Western Australia approximately 55 km south of the town of Laverton. Currently, the mine is producing from both its open pit and underground operations and is planning a significant step change in underground production as the pit phases out. The mine ventilation system is arranged in three independent ventilation districts: Astro, GQ and Cosmo. The future ventilation infrastructure requires significant upgrades to keep up with the increased tonnage. In the short term, increased production is planned for the GQ district as well as for Vogue, which is an extension in the Cosmo ventilation district. In the long term, however, Vogue is likely to become the centre of attention for mining activity.This paper explores several ventilation scenarios which include upgrading the existing fan stations, rentals, establishing a common ventilation backbone, and establishing new upcast infrastructure for GQ to support the short-term production increase. These developments were modelled using ventilation network simulation software to determine airflow distribution and fan operating points for each option. For the GQ area, with its short-term increase in production, each option is evaluated for total owning cost by considering capital and operating costs over a three-year period. For Vogue, which is to become the main focus of mining in the future, a different strategy is followed in analysing the ventilation requirements. Instead of considering different short-term options, a number of staged upgrades are considered to achieve the desired production rates.This paper summarises the planning criteria, airflow requirements, ventilation infrastructure requirements, and economic analysis associated with the conceptual ventilation system design. The current method employed to quantify the efficiency of separation of ferromagnetic ores by wet low intensity magnetic drum separators (WLIMS) incorporates the use of a conventional benchscale Davis Tube (DT) operating at a magnetic field intensity that is equivalent to that of the drum’s surface. However, empirical research has indicated that the magnetic force, or force index, rather than the magnetic field intensity, is the critical factor in the operation of the magnetic separator (Murariu and Svoboda, 2002). Furthermore, the current aforementioned practice would yield incorrect information as the magnetic field gradients produced by a typical WLIMS unit differs considerably from that of a bench-scale DT. In light of this, research on magnetite focusing primarily on the scale-up of DT results to pilotscale WLIMS were undertaken. The investigation entailed a comparative study between that of a typical WLIMS circuit and a bench-scale DT. The purpose was to practically evaluate the performance of a WLIMS, bench-marked against the traditional laboratory-scale DT unit. Moreover, the secondary objective was to produce a magnetite concentrate graded at 69% Fe, 95% -45μm, as a suitable source of separation media for use in the Dense Medium Separation (DMS) industry. Fundamental findings of previous works provided a framework for the study, however, a realworld approach was explored. Preliminary testwork conducted at Mintek investigated the performance of a DT and a typical multi-stage laboratory-scale WLIMS circuit (operating at 1000 Gauss at the surface). The results attained indicated that a DT operating at high intensities is comparable to a single rougher WLIMS stage. Additionally, it was observed that the three stage laboratory-scale WLIMS circuit performed similarly to the DT operating at 1000 Gauss. Moreover, piloting testwork was undertaken, carried out in larger 380mm drums, and the results attained showed a strong correlation with laboratory-scale findings. A low volatile medium coking coal from Jharia coal field, India was used for this investigation. The proximate analysis of the sample shows that it contains about 25% ash, 21.4% volatile matter and 52.4% fixed carbon. The sizewise ash analysis of -0.5 mm coal indicates that -0.5+0.15 mm fraction contains lower ash (21.9%) than -0.15 mm fraction (29.7%). The flotation characteristics of the -0.5 mm coal were determined by release analysis. The study reveals that recovery of combustible at 15% ash is about 27% and 43% at 17% ash level. The low recovery is due to the presence of high ash in finer fraction. In order to improve the recovery, the -0.5 mm feed was classified into -0.5+0.15 mm and -0.15 mm fractions, and subjected to flotation separately. The recoverable combustible obtained from the release analysis of -0.5+0.15 mm at 15% ash level is 46%. The effect of collector dosage, frother dosage and aeration rate on flotation was studied. The responses of these variables in collectorless flotation of the -0.5+0.15 mm fraction containing low ash were also studied. A lower aeration and frother dosage favored the generation of clean coal with low ash. The ash content in the tailing stream from -0.5+0.15 mm flotation circuit is close to that of the original -0.15 mm fraction. The former is ground to -0.15 mm and the two fractions are processed together. As -0.15 mm fraction contains relatively higher ash, collector aided flotation using sodium silicate was performed to recover the clean coal with 17% ash. The products of the overall flotation circuit having different ash levels were recommended for use in different applications. The reject from -0.15 mm flotation circuit contained 56.4% ash which can be used for fluidized bed combustion (FBC). This eventually leads to complete utilization of coal.

This paper discusses the development of a strategy for ensuring long-term geotechnical stability of final pit walls at AngloGold AustraliaÆs Sunrise Dam Gold Mine. The strategy makes use of improved blasting techniques, changes to the mining sequence and other geotechnical techniques. Only the blasting techniques will be discussed in this paper. These were developed from numerous trials over a period of several months from May 2003 through to November 2003. The focus was to reduce crest loss damage and thereby improve final wall integrity, maintain designed safe berm widths, and ensure long-term stability. Crest loss mechanisms are now understood and are explained in this paper. The preferred solution is outlined; which is a combination of improved blasting and changes in the mining sequence to cater for pre-existing ground conditions. These ground conditions include dynamic hyper saline water with sometimes intersecting steeply dipping discrete structures. Project requirements were defined as mitigating or eliminating crest loss using 127 mm diameter pre-splitting and targeting minimal production blasting vibration levels against the walls. Blast results were measured by post blast visual inspections and video. Blast design input utilised the predictive algorithms built into simulation software when attempting to minimise vibration levels against the walls. Protection of the already fired pre-split line is now achieved using small charges against the wall and timing sequences which form a mask for the later firing main charges. The pre-split spacing is now modified to suit the geology. There eight blast domains identified around the perimeter of the pit, each having varying joint orientation. In addition, blasting the bench adjacent to the berm crest has particular considerations not required by the middle and lower benches. The preferred solution was to purposefully translate this damaged top section of the pre-split bench into a planned subsequent dig zone. The design of this top æweightÆ is a direct response to the nature of the crest loss mechanism.

"This paper presents a conceptual design of an innovative tunnelling method named the Ring Pipe Jacking Method (RPJ) which is conducted by the Ring Pipe Jacking Machine (RPJM). The RPJM consists of a double ring shell which forms a ring section where small interlaced cutter heads or several micro tunnelling machines are assembled to cut soil. Similar to the conventional pipe jacking method, the RPJM also needs a thrust system, a slurry transport system, and a control system. To satisfy the need for large/super large or special section tunnels in future, the RPJM is designed to reduce sharply the excavation face to the annular section, thus enhancing the face stability. Part of the innovation is that the RPJM uses modular cutter heads or micro-TBMs which can be reused. The typical double shield shape can be easily changed by reinstalling the modular cutter heads. Modularity allows the tunnel to be cut in any cross-section shapes, ranging from a circular tunnel to a rectangular one. INTRODUCTION The demand and requirement to utilize urban underground space is rising with the rapid social development in recent years. It is necessary to reduce excavations of the urban ground surface to ensure smooth traffic in cities. Therefore the conventional open cut construction method for underground tunnels can no longer satisfy the need of practical engineering, and it is taken over by trenchless technologies .The Pipe jacking method is an important trenchless technology and is used more and more frequently to install underground pipelines, but the present pipe jacking method still has many problems. For example, it is not suitable for installing large-diameter tunnels and it also has difficulty in construction of short radius curved tunnels. This paper presents a conceptual design of an innovative pipe jacking machine named the Ring Pipe Jacking machine (RPJM) and introduces the correspondent Ring Pipe Jacking Method (RPJ) which can be applied to construct large-diameter tunnels. Furthermore, technical characteristics and superiority of the ring pipe jacking method are analyzed in this paper."

Considerable attention has been, and is being, given to the physics of water-films associated with bubble interactions at surfaces. In contrast, little information on the chemistry of such films has been reported. This communication reports a method using sum frequency vibrational spectroscopy (SFVS) to describe hydrogen bonding of interfacial water molecules in water films created with bubbles at hydrophilic and hydrophobic surfaces. The SFVS spectra of water films at hydrophilic silica surfaces differ significantly from the spectra of water at a hydrophobic silica surface. The SFVS spectra indicate that a very ordered water structure exists in water films at the hydrophilic silica surface during contact with a bubble and that the extent of hydrogen bonding increases with an increase in contact pressure. Such structural considerations as well as repulsive electrostatic and Van der Waals forces account for film stability for a bubble at the hydrophilic silica surface. In contrast, the SFVS spectra of water at a hydrophobic silica surface show a lack of hydrogen bonding and are characterized by a distinct absorption at about 3700 cm-1 similar to the spectrum of the air/water interface. These results suggest the presence of a water void, or water exclusion zone, at the hydrophobic surface, as supported by X-ray reflectivity measurements reported in the literature, by atomic force microscopy, and by the results from molecular dynamics simulations. Of course the water film at the hydrophobic surface is unstable with film thinning and rupture upon bubble contact. Under these circumstances it is shown that an attractive Van der Waals force between the bubble and the hydrophobic surface can be expected when the water exclusion zone is taken into consideration. As the thickness of the water exclusion zone increases to a thickness corresponding to the size of nano bubbles, the calculated attractive Van der Waals force increases. DURING the past few years, the requirements for steel have been raised so high that soundness is more important than ever before. The old practice was to mike steel ingots of sufficiently large sections to permit a considerable reduction of area in rolling and hammering to a given size, relying on the fact that by such rolling and hammering all cavities will be squeezed together and practically eliminated. The finished sizes, if found by inspection of the surface to be faultless, were therefore pronounced to be sound. But when such steel is ?worked up,? the manufacturer is surprised and disappointed to find that it splits, or that streaks and seams make the product unmarketable. Large sections, such as rails, beams or heavy forgings, may, within certain limits, be sufficiently strong, even if the sides of the cavities are only squeezed, and not welded together. But it is well known that, in the interior of heavy articles, under certain conditions, forging may develop small streaks into large cavities. Small sections, steel for ordnance and tubes, and numberless articles in the tool-trade, always show up the defects. It may be doubted whether cavities in steel ingots can ever be completely welded. The old theory, is, that the silver-clear blow-holes, without the blue oxide, can be welded by sufficient working. They are, however, not empty, but are filled with carbonic oxide. It is admitted that the "blue" cavities, resulting from air mechanically drawn down into the mold during the casting, can never be welded, because the oxide does not melt, and there would be no escape for it if it did. The only way in which steel can be made satisfactory for the above-named particular purposes is by having it ?dead molten." But then the trouble of piping is encountered. Best practice for the submission of samples to an internal or commercial laboratory requires quality control materials to be included for the purpose of independently and comprehensively assessing laboratory performance. These quality controls include the use of certified reference materials (CRMs) and may be blind to the laboratory. Iron ore companies purchase these materials from reputable manufacturers from stock of run-of-mine materials at the appropriate grades. The international standard, ISO 17034 (ISO, 2016), specifies the competency requirements for organisations manufacturing reference materials, and includes rigorous requirements for the assessment of homogeneity, stability and establishment of property values and their uncertainties. Homogeneity is one of the most important measures of suitability for all certified reference materials and refers to the degree to which the composition and properties of a sample are uniform throughout the batch, relative to the analytical precision of the method. More specifically, homogeneity studies of candidate reference materials should be assessed for both within sample variance and between sample variance with appropriate statistical controls. The design and application of homogeneity studies are discussed as they apply to both crushed and pulverised reference materials. Crushed certified reference materials have been utilised by the iron ore industry for a number of years and provide the user with a critical assessment of variances in sample preparation in addition to analysis. Crushed reference materials are similar in presentation to an RC sample and are generally used as discrete parcels of material submitted to the laboratory from the field. Pulverise certified reference materials miss the sample preparation step and are subsampled from th delivered sachet at the point of analysis. This paper compares the relative homogeneity performance of crushed and pulverised certified reference materials by grade across key iron ore chemical constituents. Outcomes are benchmarked to the international standard ISO 9516 (ISO, 2003) for the determination of various elements in iron ores by lithium borate fusion and X-ray fluorescence. Practical implications are discussed with reference to a case study from an iron ore laboratory in the Pilbara.

Introduction The fiftieth anniversary is, for an individual or a society, an appropriate time for a review of the past so that events and experiences may be viewed m perspective, and thereby serve as a useful guide to the future. In this review of mining geology it is proposed to set forth the situation as it was when the Institute was founded, and then trace the developments during three appropriate periods-the first from 1898 to 1918 the second from 1919 to 1939 and the third from 1939 to 1948. Mining activities were widespread in Canada in 1898 and some of the operations were considered large for their day. The total value of the metals produced in that year was $21,705,854, and in the case of five metals the output had a value in excess of one million dollars. In order of importance these were gold, silver, copper, nickel, and lead. The important producing areas were ova Scotia, Quebec, Ontario British Columbia, and Yukon. This diversification of the mining areas and the metals produced created an interest in mining throughout Canada, and was an important factor in making Canadians conscious of their mining industry. In the year 1898 geology was a recognized science. The principles of physical geology were well known, but the same cannot be said of economic geology and structural geology. Ore deposits and their occurrence had been studied chiefly in Europe. A great many important observations had been made but the theories linking these facts together were still m a formative stage. The same condition was true of structural geology. These two branches of geological science which are so important in what is now called 'mining geology', had not progressed far enough to be of practical assistance mining operations. For this reason, geologists were regarded as dreamers in ivory towers-a concept chat still lingers in the minds of those who have not kept abreast of the growth of the science.

(Pittsburg Meeting, March, 1910.) IN the copper-mining district of northern Michigan a fair-sized mine usually operates two or more shafts along the strike of the lode, these shafts being usually at least 1,000 ft. apart. The tonnage to be hoisted from each shaft is large-from 13,000 to 25,000 tons per month. The hoisting-engines used are generally steam-driven, non-condensing, duplex engines with Corliss valves, supplied by steam from separate boiler-plants where the distance between shafts is great. In an attempt to centralize power-plants and secure central-station economies, the Winona Copper Co. installed an electric hoist of the Ilgner type, with Ward Leonard control. A brief description of this plant has already been published.1 The only point of variation from the simple Ilgner set lies in the connection of two generators to a single induction-motor by flexible couplings. These generators serve motors which operate hoisting-drums at separate shafts, viz., No. 4 shaft, Winona, and No. 1 shaft, King Philip. , The hoist for No. 4 shaft, is about 1,700 ft. from the power-plant, and in this building the motor-generator set is placed. No. 1 King Philip hoist is 1,500 ft. from the motor-generator set. The equipment of the motor-generator set is as follows: In the middle of the set is a 12-pole, 450-h-p., 600-rev. per min., 2,080-volt, three-phase, 60-cycle, variable-speed induction-motor, connected on each side to a 20-ton fly-wheel which is 10 ft. in diameter, and to a 6-pole, 170-kw, 575-volt, interpole, direct-current generator. On each side of the shaft is placed a separate exciter, which delivers current at 125 volts. There is a growing interest in magnetite ores because of the increasing demand for this commodity. With these ores, there is significant cost associated with milling the ore to fine sizes using ball mills in closed circuit with hydrocyclones. Cyclone classification efficiency tends to be poor due to the large difference in density between magnetite and gangue. This decreases the grinding circuit efficiency and results in over-grinding of magnetite. The objective of the work described in this paper was to modify the Bond locked cycle milling test procedure to quantify the loss of grinding circuit efficiency due to the mineral density effect. The locked cycle test procedure described here includes a stage of magnetic separation treating the closing screen undersize, which allows simulation of magnetite classification. The effect of classifier efficiency on low-density particles (non-magnetics) was simulated by altering the proportion of closing screen undersize bypass returning to the mill. Using a magnetic separator and a screen in the magnetics stream, different proportions of the magnetic fraction can be bypassed, thus allowing simulation of the effect of particle density on classification efficiency. This laboratory test work has confirmed that the impact of mineral density on cyclone efficiency subsequently has a crucial negative effect on ball mill circuit capacity. The results are in good agreement with reductions in capacity observed in practice. Multicomponent grinding and classification models were also developed in the LIMN software environment and were able to successfully simulate both the industrial and the laboratory ball mill circuit. Loussavaara-KiirunavaaraAB?s (LKAB) iron ore mine, Kiirunavaara, is of magnetite type and associated with apatite, a calcium phosphate mineral. Rare earth elements (REE) have been found in connection with the apatite but also in other minerals. An apatite concentrate was produced by flotation, in order to investigate the possibilities to extract REE from the apatite by hydrometallurgical means. The content of rare earth elements calculated as oxides (REO) was 0.44% in the concentrate. Initially, leaching was done with large excess of sulfuric acid, hydrochloric acid, and nitric acid to study the REE solubility with different acids. In sulfuric acid, the leaching recoveries of the different REE varied between 35% and 69%. The reasons for the relative low recoveries, were that the rare earth elements precipitated together with huge amounts of gypsum, that was formed during leaching. However, both hydrochloric- and nitric acid gave almost complete extraction of the REE into solution. Analysis of the small leaching residue obtained, revealed, that mainly the light REE remained in the residue indicating that a small part of the light REE were present in the concentrate, in some acid, insoluble mineral other than apatite. With the aim to separate the REE from the phosphoric acid, the solutions from hydrochloric- and nitric acid leaching were neutralized to a pH of approximately 2 with ammonia. During neutralization, REE were precipitated as phosphates with good recoveries and a concentrate of 10.5% REO suitable for further upgrading was obtained, while the remaining solution can be used for fertilizer production. In further solvent extraction studies, it was shown, that after re-dissolution the REE could be effectively extracted by Cyanex 923, thus allowing for purification from calcium and phosphate ions. "A graphical method to assess the size reduction by grinding is presented and illustrated with data collected from laboratory grinding tests and industrial grinding circuits. The method lies between the graphical comparison of size distributions and the population balance model. The method is easy to apply and provides a quick way to compare grinding mill performances under various operating conditions and processed ores. The application of the method to industrial mills shows that a grinding mill operation could present an invariant behaviour that can be exploited for simulation purposes.INTRODUCTIONThe size reduction of particles by grinding has been studied for decades and several books were published on the subject. Most of the recent studies focus on the modelling of the ball, semiautogenous and autogenous grinding mills operation (Napier-Munn et al., 1996) because accurate simulators can be used for design, optimization and control. Although the development of realistic simulators is progressing, the trial and error method is still used by many operators to identify optimum operating conditions for a grinding machine.Laboratory tests allow the characterization of the material submitted to grinding and the analysis of various operating conditions, such as ball size or shape, mill speed and slurry concentration of solids, on particle size reduction. Similar tests can be conducted in a plant, but results are often difficult to analyze due to errors associated with sampling and sensor accuracy. This paper presents a graphical method to assess the degree of size reduction carried out by a grinding machine. The paper consists of three sections. The first section describes the method and the following sections illustrate the method with data taken from laboratory and industrial grinding machines."

Although an almost continuous campaign had been carried on by various experimenters since 1885 to find a commercial process of smelting zinc by electrothermic means, no commercial success was attained in the United States. The St. Joseph Lead Co., appreciating the theoretical possibilities of such a process, such as the ability to construct units of large capacity, quality control, and the possibility of treating complex and refractory ores, decided in 1926 that the process was worthy of further investigation, and in that year, at one of its southeast Missouri plants, began the development of the electrothermic zinc-smelting process according to the invention of the late Earl C. Gaskill. Late in 1929 the process reached a stage of commercial significance, and in 1930 the erection of a modern electrothermic zinc smelter was begun, at Josephtown, Beaver County, Pa. The site is 3 miles from Monaca, which is on the main line of the Pittsburgh & Lake Erie Railroad, and a spur of the railroad has been constructed to the plant site. The site borders upon the Ohio River, which is navigable for 600 to 800-ton barges. This district was selected for the plant after an economic survey involving such questions as cost and availability of electric power, reduction fuel, transportation facilities, and the transportation charges on raw and finished materials, labor conditions, and taxes. The initial unit of the plant was designed to treat daily 120 tons of 58 per cent zinc sulphide concentrates from the company's mines in St. Lawrence County, New York. The plant equipment is housed in seven modern steel-frame buildings, and includes: Roasting and Acid Plants 3000-ton concentrate storage. Two Nichols-Herreshoff 21 ft. 6-in. diameter, 12-hearth roasters with usual cast-iron airshaft supporting chromium-alloy rabble arms and rabbles. SO2 gas purification system consisting of: One double-compartment hot-dust Cottrell precipitator. Cast-iron sinuous air-contact gas cooler.

INTRODUCTION The goal of any development drilling program is the drilling of the exact number of holes necessary to adequately determine grade and dimensions of an orebody, no more and no less. This cost of development drilling represents a large part of the total development budget. Any technique which eliminates unnecessary drilling costs produces beneficial results, both to the company and to the person who can claim the savings. This high development cost factor has produced a quantity of research to develop techniques, for efficient control of drilling programs. Recent mining literature is full of articles describing methods for measuring and increasing the effectiveness of these expensive projects. The method described in this paper was developed during the latter part of a major development drilling project at the Christmas Mine of Inspiration Consolidated Copper Company in Gila County, Arizona. It was used to test the need for drilling individual holes but could be expanded to control an entire project. This technique was used at Christmas as a practical means for evaluating adequacy of drilling on an individual hole basis, and it was possible to eliminate several previously approved holes. Application of this method involves an evaluation of geological conditions present and a good bit of common sense. Projects involving large disseminated, rather homogeneous orebodies of the porphyry-copper type would be the most likely candidate for application of the technique. Discontinuous orebodies would not be amenable to this technique. The method is based on the measurement of the linear correlation between the spatial distribution of two sets of data. The Pearson product-moment correlation coefficient is used. This is a standard statistic commonly used to measure linear correlation between two sets of data. The statistical significance of the correlation coefficient at a 95% confidence level is the main criterion for controlling the project. For purposes of reference we will call our technique the "correlation method" in this paper.

"This paper discusses the Advanced Technology Program (ATP)'s support over the last decade to the development and use of process modeling as a tool to address some of the challenges faced by the U.S. metal casting industries. A particular focus will be given to highlight how process modeling can be used to improve the manufacturability of components needed for high performance and critical applications. The challenges associated with the design and production of thin-walled, cast parts with intricate and complex internal geometries will be discussed. Modeling issues associated with the control of the final grain structure in the solidified parts will also be addressed. This paper will describe the advantages of using appropriate mathematical models for designing new alloys for use in various applications.Finally, the role that ATP has played in uplifting the U.S. technological base through partnerships with industry and academia in high-risk research in order to bring to market new and improved products for the broader benefit of the nation will be discussed.ATP's BackgroundIn the 1980s, U.S. industries were losing market share to low-cost, high-quality manufacturers from Asian countries. Corporate leaders and policy makers determined that in order to sustain and grow market share, U.S. industries needed to improve their competitiveness and productivity through product and process innovation. There was evidence that companies were systematically under-investing in leading-edgetechnologies and failing to complete the development of new technologies [l]. There were also arguments in academic circles that pointed to a potential market failure in the area of early-stage technological developments. These concerns provided impetus for creation of the Advanced Technology Program (ATP) within the National Institute of Standards and Technology through the Omnibus Trade and Competitiveness Act of 1988. Through a rigorously competitive, cost-shared partnership with industry, ATP fosters the development and broad dissemination of high-risk technologies that offer the potential for significant broad-based economic benefits to the United States."

At the Twin Cities "lining Research Center (TCMRC) a project is underway to determine how varying the parameters of a coal-cutting process would affect the amount and the size distribution of the generated dust. The cutting parameters being investigated are the cutting speed, depth of cut and shape of the cutting, tool. To accomplish this research ft is necessary to collect and transport the coal dust venerated at the cutting tool to measuring instruments for analysis. The purpose of this contract was to develop and deliver a system of instruments for measuring the size distributions of the generated coal dust. The instruments employed are essentially those developed for particulate air pollution measurements (1, 2). The instrument system covers a particle size range of 0.005 to 20 microns and is capable of performing in situ measurement and presenting data in real time.

"A sensitive colorimetric assay for detecting mucins and glycoproteins rich in 0-glycosidically linked saccharides is reported. The method combines the susceptibility of N-acetylgalactosamine terminating 0-glyco idically linked saccharides to P-elimination with the Morgan-Elson reaction for N-acetylhexosamines with free reducing ends. All mucin and mucin-type glycoproteins but none of the serum-type glycoproteins tested resulted in characteristic color production. All mucins tested gave linear responses in the range 5 to 200 ""g and the assay was also adapted to the microscale involving the use of 96-well microtiter plates. The microassay in which the volumes of samples and reagents are scaled down 2.5-fold was particularly useful in monitoring of mucins, in the presence of other glycoconjugates, in large numbers of samples obtained during fractionation procedures. Cesium chloride, cesium bromide, potassium thiocyanate, and various detergents do not interfere with the colorimetric determination. Guanidine hydrochloride, cesium triftuoroacetate, and B-mercap- . toethanol decreased color by 30 to 45%; however, the interference was not serious to prevent the use of the method for detection of mucins in their presence. The use of the method for the specific detection of mucin during fractionation by gel filtration and density gradient centrifugation of cystic fibrosis sputum samples is demonstrated. Mucins are an important class of biomacromolecules whose structure-function relationship is poorly understood (1,2). The investigations of mucins are hampered by difficulties in purifying these very large size molecules which appear to avidly bind other components of the mucus (3,4). One of the problems during purification is the specific detection of mucins. For example, during the various purification steps, the fractions containing mucins are typically detected by assaying for neutral sugars (hexoses) (5,6), for sialic acids (7), or for periodate oxidizable saccharide units (8). However, these methods are not entirely satisfactory because neutral sugars, sialic acids, and periodate-oxidizable saccharides are also components of other glycoconjugates. In addition, the neutral sugar and sialic acid measurements are also subject to interference from high levels of proteins and nucleic acids."
